大鼠局灶性脑梗死后大脑皮质PRX6表达和MDA含量的变化

摘要: 目的: 观察局灶性脑梗死大鼠梗死区大脑皮质过氧化物酶6(peroxiredox6,PRX6)表达和丙二醛(malondialdehyde,MDA)含量的变化。方法: 30只雄性SD大鼠随机分为对照组和模型组,采用线栓法制作大鼠大脑中动脉栓塞模型,各组大鼠分别于术后1d、3d、7d处死取材。采用硫代巴比妥酸法检测梗死区大脑皮质MDA的含量,Western blot法和qRT-PCR法检测梗死区大脑皮质PRX6蛋白和mRNA的表达。结果: 模型组各时间点大鼠梗死区大脑皮质MDA含量,PRX6蛋白、mRNA表达均明显高于对照组(P<0.01)。随着术后时间的延长,模型组大鼠梗死区大脑皮质MDA含量逐渐降低,PRX6蛋白、mRNA表达逐渐升高(P<0.01)。结论: 大鼠脑梗死后可能通过上调PRX6的表达水平减轻氧化应激损伤。

Abstract: Objective: To investigate the changes of peroxiredox6 (PRX6) expression and malondialdehyde (MDA) content in cerebral cortex of rats after local cerebral infarction. Methods: 30 male SD rats were randomly divided into control group and model group. Suture method was used to establish middle cerebral artery occlusion rats’ model, and the rats were killed respectively 1d, 3d, 7d after operation. Thiobarbituric acid method was used to detect the MDA content in cerebral cortex, Western blot and qRT-PCR to detect PRX6 protein and mRNA expression in cerebral cortex. Results: The MDA content and PRX6 expression in infarct area of cerebral cortex of rats in model group at each time point were obviously higher than control group (P<0.01). With time extension after operation, the MDA content declined gradually, while the PRX6 expression increased gradually (P<0.01). Conclusions: After cerebral farction, the rats may reduce oxidative stress injury by upregulating PRX6 expression.
