茵栀黄对结构性雄甾烷受体CAR介导CYP3A4的转录调节作用

TRANSCRIPTIONAL REGULATION OF YINZHIHUANG ON CYP3A4 MEDIATED BY STRUCTURAL MALE STEROID RECEPTOR CAR

摘要: 目的: 探讨茵栀黄对CYP3A4转录表达的影响及机制。方法: Chang Liver细胞中加入0.01、0.1和1μM茵栀黄分别作用24h、36h和48h,采用RT-PCR法和报告基因方法检测CYP3A4 mRNA的表达水平。结果: 不同浓度茵栀黄分别处理Chang Liver细胞24h和36h可明显升高CYP3A4 mRNA的表达水平(P<0.05);转入CAR载体的Chang Liver细胞经不同浓度茵栀黄处理36h均可激活CYP3A4的转录表达。结论: 茵栀黄可能通过CAR介导增加Chang Liver细胞CYP3A4的转录表达。

Abstract: Objective: To investigate the effects and mechanisms of Yinzhihuang on transcription of CYP3A4. Methods: The Chang Liver cells were cultured with 0.01μM, 0.1μM, 1μM Yinzhihuang respectively for 24h, 36h and 48h. The expression of CYP3A4 mRNA was detected by RT-PCR and dual-luciferase reporter assay. Results: The Chang Liver cells were cultured with Yinzhihuang of different concentration for 24h and 36h could obviously increase the expression of CYP3A4 mRNA (P<0.05). The Chang Liver cells that transfected CAR vector were cultured with different concentrations of Yinzhihuang for 36h could activate transcription of CYP3A4. Conclusions: Yinzhihuang may increase the transcriptional expression of CYP3A4 in Chang Liver cells mediated by CAR.
