17-AAG对人绒毛膜癌JAR细胞MMP-2和MMP-9表达的调控作用*

摘要: 目的: 探讨热休克蛋白90(HSP90)抑制剂17-丙烯胺基-17-去甲氧基格尔德霉素(17-AAG)对人绒毛膜癌JAR细胞基质金属蛋白酶(MMP)-2、MMP-9蛋白表达的调控作用。方法: 体外培养人绒毛膜癌JAR细胞,以不同浓度的17-AAG作用JAR细胞,采用MTT法检测JAR细胞的增殖抑制率,Western blotting法检测MMP-2、MMP-9蛋白的表达情况。结果: MTT结果显示,17-AAG对JAR细胞生长具有明显的抑制作用,并呈浓度依赖性。5μg/ml、10μg/ml、20μg/ml、40μg/ml的17-AAG作用后,JAR细胞MMP-2、MMP-9蛋白的表达明显下调。结论: 17-AAG对人绒毛膜癌JAR细胞的增殖具有明显的抑制作用,且能抑制JAR细胞MMP-2、MMP-9蛋白的表达。

Abstract: Objective: To explore the effects of heat shock protein 90 inhibitor-17-AAG on matrix metalloproteinase-2 (MMP-2) and MMP-9 expression in human choriocarcinoma JAR cells. Methods: The human choriocarcinoma JAR cells were cultured in vitro and treated with 17-AAG in different concentration. MTT was used to measure the proliferation inhibiting rate of JAR cells, Western blotting to detect the MMP-2 and MMP-9 expression. Results: MTT method showed that 17-AAG can obviously inhibit growth of JAR cells, and in concentration dependent manner. After the JAR cells were treated with 17-AAG in different concentration (5μg/ml, 10μg/ml, 20μg/ml, 40μg/ml), the MMP-2 and MMP-9 expression in JAR cells down regulated obviously. Conclusions: 17-AAG can significantly inhibit the proliferation of human choriocarcinoma JAR cells, and can inhibit the expression of MMP-2 and MMP-9 in JAR cells.
