维生素D调节多囊卵巢综合征大鼠的炎症状态

Vitamin D Regulates Inflammatory State in Rats with Polycystic Ovary Syndrome

摘要: 目的 探讨维生素D对多囊卵巢综合征(PCOS)大鼠炎症状态的影响。方法 24只3周龄雌性SD大鼠随机分为对照组、PCOS组、VD治疗组(PCOS+VD),每组8只。后2组每日灌胃1mg/kg来曲唑,高脂饲料喂养,对照组灌胃同体积0.5%羧甲纤维素钠,普通颗粒饲料喂养。连续28d后,PCOS+VD组每日灌胃200IU/kg VD,对照组和PCOS组灌胃同体积玉米油,普通颗粒饲料喂养。给药21d后取血及卵巢。ELISA测定血清 25-(OH)D、睾酮(T)、黄体生成素(LH)、促卵泡激素(FSH)、白细胞介素-6(IL-6)、白细胞介素-17(IL-17)、肿瘤坏死因子-(TNF-α)、c反应蛋白(CRP)、空腹血糖(FPG)、空腹胰岛素(FINS),western blot检测卵巢组织IL-6、IL-17、TNF-α、CRP蛋白,HE染色观察卵巢组织形态学变化。结果 PCOS组血清25-(OH)D较对照组降低(P<0.05),血清T、LH、FPG、FIN水平均升高(P<0.05),血清及卵巢组织IL-6、IL-17、TNF-α、CRP含量均升高(P<0.05),动情周期紊乱,卵巢呈多囊样改变。PCOS+VD组血清25-(OH)D升高,T、LH、IL-6、IL-17、TNF-α、CRP、FPG、FIN均降低,除血清TNF-α外,差异均有统计学意义,动情周期紊乱得到改善,颗粒细胞层数增多,排列整齐,囊状卵泡数量减少。结论 VD降低PCOS大鼠炎症因子含量,改善炎症状态。

Abstract: Objective To investigate the effects of vitamin D on inflammatory state in rats with polycystic ovary syndrome (PCOS). Methods Twenty-four immature female SD rats(three weeks-old, 57±6g)were randomly divided into control, PCOS, and PCOS+VD group, 8 rats in each group. In the last 2 groups, letrozole (1mg/kg·d, in 0.5% carboxymethyl cellulose) and high-fat diet were administered intragastrically, meanwhile, control rats were intragastric with solvent. Twenty-eight consecutive days later, the PCOS+VD group was intragastric with 200IU/kg·d VD daily, the control group and PCOS groups were intragastric with corn oil at the same volume. ELISA was performed to detect serum 25-(OH)D, T, LH, FSH, IL-6, IL-17, TNF-α, CRP, FINS, and IL-6, IL-17, TNF-α, CRP proteins of ovarian tissue were measured by western blot analysis. HE staining was carried out to observe the ovarial morphological changes. Results Serum 25-(OH)D level in PCOS rats decreased (P<0.05), while serum T, LH, FPG, FINS level increased(P<0.05), and the level of IL-6, IL-17, TNF-α and CRP significantly increased in both serum and ovarian tissue compared with control rats(P<0.05). The estrous cycle of the PCOS rats was disturbed, and the ovary presented a large number of cystic follicles, with sparse corpus luteum. Serum 25-(OH)D in PCOS+VD rats was significantly higher than that in PCOS rats. However, the level of T, LH, IL-6, IL-1,7 TNF-α, CRP, FPG and FIN was reverse, and the estrous cycle was relatively regular with fewer cystic follicles and more corpus lutea. Conclusion VD can reduce the content of inflammatory factors and improve the inflammatory state in PCOS rats.
