柑橘皮提取物对脂多糖诱导的大鼠肺部炎症及氧化应激的影响

Effect of Citrus reticulata Blanco Extract on Lung Inflammation and Oxidative Stress in Rats Induced by Lipopolysaccharide

摘要: 目的 探讨柑橘皮提取物(CRE)对脂多糖(LPS)诱导的大鼠肺部炎症及氧化应激的影响。方法 向成年雄性Wistar大鼠腹腔注射1 mg/kg/d的LPS(14 d)来诱导肺部炎症。将大鼠随机分为5组:对照组(Control组)、LPS组、LPS+CRE 100 mg/kg组(LPS+CRE100组)、LPS+CRE 200 mg/kg组(LPS+CRE200组)、LPS+CRE 500 mg/kg组(LPS+CRE500组),应用不同剂量的CRE(100、200和500 mg/kg)进行联合治疗(14 d)。检测各组大鼠的血清细胞计数以及支气管肺泡灌洗液(BALF)的细胞因子和氧化应激生物标志物水平。通过苏木精和伊红(H&E)染色评价大鼠的肺部形态。结果 CRE降低了LPS大鼠血清白细胞总数以及中性粒细胞、单核细胞和嗜酸性粒细胞百分比(P<0.05)。CRE治疗可剂量依赖性地降低LPS大鼠BALF中的丙二醛(MDA)水平,但显著提高氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性(P<0.05)。CRE治疗可剂量依赖性地增加LPS大鼠BALF中的白介素-4(IL-4)水平,但显著降低干扰素-γ(INF-γ)、转化生长因子-β1(TGF-β1)和前列腺素E2(PGE2)的水平(P<0.05)。用CRE治疗可显著降低LPS给药引起的肺组织病理学改变。结论 CRE通过降低LPS诱导的大鼠血清中白细胞总数以及中性粒细胞、单核细胞和嗜酸性粒细胞百分比,并且抑制肺部过度的氧化应激和炎症反应,从而发挥了良好的抗肺部炎症的作用。

Abstract: Objective In order to investigate the effects of Citrus reticulata Blanco peel extract (CRE) on lipopolysaccharide (LPS)-induced pulmonary inflammation and oxidative stress in rats. Methods In this study, adult male Wistar rats were intraperitoneally injected with 1 mg/kg/d of LPS (14 d) to induce lung inflammation. The rats were randomly divided into 5 groups: control group (Control group), LPS group, LPS + CRE 100 mg / kg group (LPS + CRE100 group), LPS + CRE 200 mg / kg group (LPS + CRE200 group), LPS + CRE 500 mg / kg group (LPS + CRE500 group). Various doses of CRE (100, 200, and 500 mg/kg) were administered in combination therapy (14 d). Serum cell counts and cytokine and oxidative stress biomarker levels of bronchoalveolar lavage fluid (BALF) were measured in each group of rats. The lung morphology of the rats was evaluated by hematoxylin and eosin (H&E) staining. Result CRE decreased the total number of serum leukocytes and the percentage of neutrophils, monocytes, and eosinophils in LPS rats (P<0.05). CRE treatment dose-dependently reduced MDA levels in BALF in LPS rats, but significantly increased SOD and CAT activity (P<0.05). CRE treatment dose-dependently increased the levels of IL-4 present in the BALF of LPS rats, but significantly decreased the levels of INF- γ, TGF- β1, and PGE 2 (P<0.05). Treatment with CRE can significantly reduced the lung histopathological changes induced by LPS administration. Conclusion CRE exerted good effects against pulmonary inflammation by reducing the total number of leukocytes and the percentage of neutrophils, monocytes and eosinophils in rat serum induced by LPS, and by inhibiting excessive oxidative stress and inflammation in the lungs.
