摘要/Abstract

摘要： 目的探讨原肌球蛋白调节蛋白1（Tmod1）过表达对大鼠血管平滑肌细胞迁移和侵袭的影响。方法采用酶消法提取原代大鼠胸主动脉血管平滑肌细胞,通过免疫荧光SM22α染色方法鉴定血管平滑肌细胞。使用腺病毒感染的方法,将Tmod1的腺病毒感染到血管平滑肌细胞中,对照腺病毒Ad-Null感染对照组细胞。培养48 h后,用蛋白质印迹法(Western blot)鉴定各组Tmod1的过表达效率,细胞划痕实验和Transwell实验比较过表达Tmod1组细胞与对照组细胞的迁移能力和侵袭能力;RT-qPCR检测Tmod1过表达后基质金属蛋白酶15(MMP15)表达水平变化。结果过表达Tmod1后,大鼠血管平滑肌细胞的迁移和侵袭能力均显著增加(P<0.05),MMP15的表达显著上升(P<0.05)。结论 Tmod1过表达可能是通过靶向调控MMP15的表达来促进血管平滑肌细胞迁移和侵袭的能力。

Abstract: Objective To investigate the effect of tropomodulin 1 (Tmod1) overexpression on the migration and invasion of rat vascular smooth muscle cells (VSMCs). Methods Rat vascular smooth muscle cells were cultured and identified by immunofluorescence SM22a staining. Adenovirus encoding Tmod1 was used to infect VSMCs and control adenovirus Ad-null,was used to infect control VSMCs. After 48 hours of culture, the expression of Tmod1 in each group was detected by Western blot to identify the overexpression efficiency. The cell scratch test and Transwell assay can detect the fuction of cell migration. Rt-qpcr was used to detect the expression of matrix metalloproteinase MMP15 after Tmod1 overexpression. Results Overexpression of Tmod1 promoted the migration and invasion of rat vascular smooth muscle cells (P<0.05), and the expression of MMP15 was significantly increased (P<0.05). Conclusion Tmod1 overexpression may promote cell migration and invasion possibly by targeting MMP15 expression.
