碘普罗胺对心室肌细胞离子通道的影响

摘要: 目的:观察非离子型低渗性造影剂碘普罗胺对心室肌细胞离子通道的影响。方法:急性分离大鼠心室肌细胞,并使用全细胞膜片钳技术记录碘普罗胺干预前后钠离子电流(INa)、钙离子电流(ICa,L)、瞬时外向钾离子电流(Ito)的电流密度、稳态激活曲线、失活曲线的改变。结果:与碘普罗胺干预前相比,碘普罗胺干预后ICa,L和INa的电流密度降低,Ito电流密度升高;而碘普罗胺对ICa,L、INa和Ito的稳态激活和失活曲线均无明显影响。结论:碘普罗胺可能通过改变心室肌细胞动作电位的时程导致恶性室性心律失常的发生。

Abstract: Objective: To observe the effects of non-ionic hypoosmotic contrast agent, iopromide on ion channels in ventricular myocytes. Methods: The ventricular myocytes were isolated and whole cell patch-clamp technique was used to record the current density, steady-state activation curve and inactivation curve of sodium current (INa), L-type calcium current (ICa,L) and instantaneous outward potassium ion current (Ito) before and after iopromide administration. Results: Compared with that before iopromide administration, the current density of INa and ICa,L decreased obviously, the current density of Ito increased obviously after iopromide administration. While iopromide has no obviously effects on steady-state activation curve and inactivation curve. Conclusions: Iopromide may induce malignant ventricular arrhythmia by changing the duration of action potential of ventricular myocytes.
