miR-15a-5p和MDR1在大肠癌组织中的表达及临床意义*

摘要: 目的:研究miR-15a-5p和多药耐药基因1(MDR1)mRNA在大肠癌组织中的表达及意义。方法:采用实时荧光定量PCR法检测44例大肠癌组织及对应的癌旁正常肠黏膜组织中miR-15a-5p、MDR1 mRNA的表达情况,分析miR-15a-5p、MDR1 mRNA表达与大肠癌临床病理特征的关系,以及大肠癌组织中miR-15a-5p与MDR1 mRNA表达的相关性。结果:大肠癌组织miR-15a-5p的相对表达量明显低于癌旁正常组织,MDR1 mRNA的相对表达量明显高于癌旁正常组织(P<0.05)。大肠癌组织miR-15a-5p、MDR1 mRNA表达均与TNM分期和淋巴结转移有关(P<0.05)。Spearman相关分析显示,大肠癌组织miR-15a-5p表达与MDR1 mRNA表达呈负相关关系(r=-0.343,P<0.05)。结论:大肠癌组织中miR-15a-5p呈低表达、MDR1呈高表达,二者可能在大肠癌的发生发展过程中发挥重要作用。

Abstract: Objective: To study the expression and clinical significance of miR-15a-5p and multidrug resistance gene 1(MDR1) mRNA in colorectal carcinoma tissues. Methods: RT-qPCR was used to detect the expression of miR-15a-5p and MDR1 mRNA in 44 cases of colorectal carcinoma and adjacent non-tumor tissues. The relationships between miR-15a-5p, MDR1 mRNA and clinical pathological features of colorectal carcinoma, as well as the correlations between miR-15a-5p and MDR1 mRNA in colorectal carcinoma were also analyzed. Results: Compared with the adjacent non-tumor tissues, miR-15a-5p mRNA expression was significantly lower in colorectal carcinoma, but MDR1 mRNA expression was significantly higher (P<0.05). The miR-15a-5p and MDR1 mRNA expression were all concerned with lymph node metastasis and TNM stage of colorectal carcinoma (P<0.05). In colorectal carcinoma, miR-15a-5p mRNA expression was negatively correlated with MDR1 mRNA expression (r=-0.343, P<0.05). Conclusions: The expression of miR-15a-5p was low and MDR1 was high in colorectal cancer tissues. Both of them may play an important role in development of colorectal carcinoma.
