INHIBITORY EFFECTS OF GANODERMA LUCIDUM POLYSACCHARIDES ON VEGF SECRETION IN B16F10 MELANOMA CELLS

灵芝多糖对小鼠黑素瘤B16F10细胞分泌VEGF的抑制作用

Abstract: Objective: To investigate the effects of Ganoderma lucidum polysaccharides (GL-PS) on secretion of vascular endothelial growth factor (VEGF) in B16F10 melanoma cells. Methods: B16F10 melanoma cells were treated with or without different concentrations of GL-PS for 48h, then the expression of VEGF protein and mRNA was respectively detected by Western-blot and RT-PCR. Results: Compared with control group, the expression of VEGF protein and mRNA in B16F10 melanoma cells decreased obviously after the B16F10 melanoma cells were treated with different concentrations of GL-PS (0.8μg/ml, 3.2μg/ml, 12.8μg/ml) for 48h (P<0.05). Conclusions: GL-PS may play a role in inhibiting tumor by inhibiting secretion VEGF in B16F10 melanoma cells.

摘要: 目的: 探讨灵芝多糖对B16F10黑素瘤细胞分泌血管内皮生长因子(VEGF)的影响。方法: 用不同浓度的灵芝多糖处理B16F10黑素瘤细胞,以正常培养基代替灵芝多糖培养B16F10黑素瘤细胞为对照。分别采用Western blot法和RT-PCR法检测灵芝多糖作用48h后B16F10黑素瘤细胞VEGF蛋白和mRNA表达的变化。结果: 浓度为0.8μg/ml、3.2μg/ml、12.8μg/ml的灵芝多糖处理B16F10细胞后,B16F10细胞VEGF蛋白和mRNA的表达水平均明显降低(P<0.05)。结论: 灵芝多糖可能通过抑制B16F10黑素瘤细胞分泌VEGF起到抑制肿瘤的作用。
