原肌球调节蛋白1对小鼠肾脏尿浓缩功能的影响*

Abstract: Objective: To analyze the effects of tropomodulin1 (Tmod1) on urine concentration in kidney of mice. Methods: Tubule-specific Tmod1 knockout mice, Tmod1flox/flox/Ksp-cre+ (TFK) and its control mice Tmod1flox/flox/Ksp-cre-(TF)were tanken as research object in this study. The urine of mice was collected and the 24-hour urine volume as well as urine osmolality of all the mice were measured at basic state, water deprivation and water loading using metabolic cage system. Results: At basic state, water deprivation and water loading, the 24-hour urine volume of TFK mice was remarkably lower than TF mice, while the urine osmolality of TFK mice was remarkably higher than TF mice (P<0.05). Conclusions: Tmod1 may be involved in the process of urine concentration in mice, but the mechanisms need to be further studied.

摘要: 目的:分析原肌球调节蛋白1(tropomodulin1,Tmod1)对小鼠肾脏尿浓缩功能的影响。方法:采用肾脏集合管和肾小管特异性敲除Tmod1小鼠Tmod1flox/flox/Ksp-Cre+(TFK)及其对照小鼠Tmod1flox/flox/Ksp-Cre-(TF)为研究对象,利用代谢笼系统,分别在基础状态,以及给予小鼠禁水和水负荷处理,收集小鼠尿液测量小鼠24小时尿量和渗透压。结果:基础状态,禁水和水负荷条件下,TFK小鼠的尿量均明显少于TF小鼠,尿液渗透压明显高于TF小鼠,差异具有统计学意义(P<0.05)。结论:Tmod1可能参与了小鼠尿浓缩的过程,但机制有待深入研究。
