Abstract: Objective: To investigate the regulatory effects of matrix stiffness on the elastic modulus and cytoskeleton of macrophages. Methods: Mouse macrophage RAW264.7 were cultured on different stiffnesses of polyacrylamide gels (2kPa and 20kPa). The nano-indenter was used to detect the elastic modulus of macrophages cultured on different matrix stiffness gels, flow cytometry to detect the F-actin content in macrophages. Results: Compared with 2kPa matrigel, elastic modulus and F-actin content of macrophages that cultured on 20kPa matrigel increased obviously (P<0.05). Conclusions: Matrix stiffness can regulate the elastic modulus and cytoskeleton of macrophages, but the mechanism needs to be further studied.

摘要: 目的 探讨基质刚度对巨噬细胞弹性模量和细胞骨架的调控作用。方法 制备不同刚度的聚丙烯酰胺凝胶(刚度分别为2kPa、20kPa),将小鼠巨噬细胞Raw264.7分别接种在不同刚度的基质胶上进行培养。采用纳米压痕仪检测不同刚度基质胶上培养的巨噬细胞的弹性模量,流式细胞仪检测巨噬细胞F-actin的含量。结果 与2kPa的基质胶相比,在20kPa基质胶上培养的巨噬细胞,弹性模量和F-actin的含量均显著增加(P<0.05)。结论 基质刚度对巨噬细胞的弹性模量和细胞骨架有调控作用,但机制有待深入研究。
