Effect of Mangiferin on Postmenopausal Osteoporosis and Inflammation in Rats

芒果苷对大鼠绝经后骨质疏松症的疗效及对炎症的作用

Abstract: Objective To investigate the effect of mangiferin on postmenopausal osteoporosis and inflammation in rats. Methods Sixty female SD rats were randomly divided into control group, model group and mangiferin small (50 mg·kg-1), medium (100 mg·kg-1) and large (200 mg·kg-1) dose groups, with 12 rats in each group. The osteoporosis model was established in both the model group and the experimental group according to bilateral ovariectomy, while only the adipose tissue near the ovary was removed in the control group. The expressions of bone mineral density, bone microstructure, bone metabolism indexes, inflammatory factors and related proteins were observed in each group. Results Compared with the control group, bone mineral density, bone trabecular (TB) number and bone trabecular thickness of the model group were significantly decreased, bone trabecular separation was significantly increased, serum PINP, BGP, ALP levels were significantly decreased, TNF-α, IL-6, IL-8 levels were significantly increased, NF-κB, MMP9 mRNA expression levels were significantly increased (P<0.05). Compared with model group, BMD, bone trabecular number, bone trabecular thickness, PINP level, TNF-α and IL-8 levels, NF-κB and MMP9 mRNA levels in mangiferin low-dose, medium-dose and high-dose groups were significantly increased (P<0.05). The level of bone trabecular separation in mangiferin medium dose group and high dose group was significantly decreased, the level of BGP and ALP was significantly increased, and the level of IL-6 was significantly decreased (P<0.05). Conclusion Mangiferin has a good effect in postmenopausal osteoporosis rats, which may be related to the inhibition of inflammatory response and inflammatory signaling pathway (NF-κB/MMP9).

摘要: 目的 探讨芒果苷(MF)对大鼠绝经后骨质疏松(PMOP)的疗效对炎症的作用。方法 将60只雌性SD大鼠随机分为对照组、模型组及MF小(50 mg·kg-1)、中(100 mg·kg-1)、大(200 mg·kg-1)剂量组,每组12只。模型组与实验组大鼠造模根据双侧卵巢摘除法建立骨质疏松模型,对照组仅将卵巢附近的脂肪组织切除。观察各组大鼠骨密度(BMD)、骨微结构、骨代谢指标、炎症因子以及相关蛋白的表达。结果 与对照组相比,模型组大鼠BMD、骨小梁数量、骨小梁厚度显著降低,骨小梁分离度显著上升,血清PINP、BGP、ALP水平均显著降低,TNF-α、IL-6、IL-8水平显著升高,NF-κB、MMP9的mRNA表达水平均显著升高(P<0.05)。与模型组相比,MF小剂量组、中剂量组、大剂量组BMD、骨小梁数、骨小梁厚度均显著升高,PINP水平显著上升,TNF-α、IL-8水平显著下降,NF-κB、MMP9的mRNA水平显著下降(P<0.05)。MF中剂量组、大剂量组骨小梁分离度水平均明显下降,BGP、ALP水平均明显升高,IL-6水平显著降低(P<0.05)。结论 MF在绝经后骨质疏松大鼠中具有较好的效果,可能与抑制炎症反应以及炎性信号通路(NF-κB/MMP9)有关。
