Changes in SMAD4, SOX2, and P53 in Cervical Cancer Tissues and Their Clinical Significance

SMAD4、SOX2、P53在宫颈癌组织中的变化及其临床意义

Abstract: Objective To investigate the expression changes of Smad homolog 4 (SMAD4), sex-determining region Y-box protein 2 (SOX2), and P53 gene (P53) in cancer tissues of patients with cervical cancer (CCA), as well as their predictive value for prognosis. Methods A total of 82 patients with CCA admitted to Luohe Central Hospital from January 2021 to January 2022 were retrospectively selected, serving as the observation group, and another 82 patients with benign tumors were selected as the control group. The expression levels of SMAD4, SOX2, and P53 were compared between the two groups with different pathological characteristics. The correlation between each indicator and the pathological characteristics of CCA was analyzed, and the relationship between these indicators and the prognosis of CCA patients was explored. Results The expression levels of SOX2 and P53 in the observation group were higher than those in the control group, while the expression level of SMAD4 was lower (P<0.05). The expression levels of SOX2 and P53 in patients with stages Ⅰ-Ⅱ were lower than those in patients with stages Ⅲ-Ⅳ, and those in high-differentiated patients were lower than those in moderately and low differentiated patients. The expression level of SMAD4 was higher in patients with stages Ⅲ-Ⅳ than in patients with stages Ⅰ-Ⅱ, and higher in well-differentiated patients than in moderately and poorly differentiated patients (P<0.05). The expression of SOX2 and P53 in CCA patients was positively correlated with TNM staging and negatively correlated with differentiation degree, while the expression of SMAD4 was negatively correlated with TNM staging and positively correlated with differentiation degree (P<0.05). The 2-year survival rate of patients with positive SOX2 and P53 expression was lower than that of patients with negative expression, while the 2-year survival rate of patients with positive SMAD4 expression was higher than that of patients with negative expression (P<0.05). Conclusion In CCA tissues, there is an abnormal increase in the expression of SOX2 and P53, and an abnormal decrease in the expression of SMAD4. These three indicators are closely related to the occurrence, TNM staging, and differentiation degree of CCA, and can provide effective references for clinical prognosis prediction.

摘要: 目的 探究Smad同源物4(SMAD4)、性别决定区Y框蛋白2(SOX2)、P53基因(P53)在宫颈癌(CCA)患者癌组织中的表达变化及其对预后的预测价值。方法 回顾性选取2021年1月~2022年1月漯河市中心医院收治的CCA患者82例为观察组,另选取良性肿瘤患者82例为对照组,对比2组不同病理特征下SMAD4、SOX2、P53的表达,分析各指标与CCA病理特征的相关性,并探讨其与CCA患者预后的关系。结果 观察组SOX2、P53表达情况高于对照组,SMAD4表达情况低于对照组(P<0.05);Ⅰ~Ⅱ期患者SOX2、P53表达情况低于Ⅲ~Ⅳ期,高分化患者低于中、低分化,SMAD4表达情况高于Ⅲ~Ⅳ期,高分化患者高于中、低分化(P<0.05);CCA患者SOX2、P53表达情况与TNM分期呈正相关,与分化程度呈负相关,SMAD4表达情况与TNM分期呈负相关,与分化程度呈正相关(P<0.05);SOX2、P53表达阳性患者2年生存率低于阴性患者,SMAD4表达阳性患者2年生存率高于阴性患者(P<0.05)。结论 CCA组织中SOX2、P53表达异常升高,SMAD4表达异常降低,三项指标与CCA发生、TNM分期、分化程度密切相关,可为临床预后预测提供有效参考。
