Effect of Hypoxia on the Expression of Metastasis-related Genes in Pancreatic Cancer PANC-1 Cells

缺氧对胰腺癌细胞转移和侵袭相关基因表达的影响

Abstract: Objective To investigate the effects of hypoxia on the expression of E-cadherin, Vimentin, MMP-2 and MMP-9 in pancreatic cancer PANC-1 cells. Methods Pancreatic cancer PANC-1 cells were divided into control group and hypoxic group. The hypoxic conditions were 5% CO2, 2% O2 and 93% N2 while the control condition is 5% CO2 and 95% O2. The expression levels of E-cadherin, Vimentin, MMP-2 and MMP-9 were detected by qRT-PCR and Western blot. Results The E-cadherin protein expression of control group and hypoxia group were 0.859±0.087 and 0.321±0.033 respectively; the Vimentin protein were 0.453±0.051 and 1.128±0.169 respectively; E-cadherin mRNA were 1.125±0.092 and 0.352±0.036 respectively; Vimentin mRNA were1.253±0.130 and 2.875±0.366 respectively. All the differences were statistically significant (P<0.05); The MMP-2 protein and mRNA level in the control and hypoxia groups were 0.254±0.031 and 0.839±0.073, 1.230±0.125 and 4.369±0.368 respectively and the difference was statistically significant (P<0.05); The MMP-9 protein and mRNA in the control and hypoxia groups were 0.268±0.039and 0.733±0.075, 1.452±0.023and 4.136±0.569. The difference was statistically significant (P<0.05). Conclusion Hypoxia can promote the invasion and migration of pancreatic cancer cells by regulating the metastasis-related genes.

摘要: 目的 探讨缺氧对胰腺癌PANC-1细胞侵袭性和转移性相关基因表达的影响。方法 培养胰腺癌PANC-1细胞,将其分为对照组和缺氧组,对照组常规培养于5%CO2、95%O2条件,缺氧组培养于5%CO2、2%O2和93%N2条件。采用Western blot和qRT-PCR方法检测E-cadherin、Vimentin、MMP-2、MMP-9的表达水平。结果 对照组和缺氧组E-cadherin蛋白表达量分别为0.859±0.087和0.321±0.033;Vimentin蛋白表达量分别为0.453±0.051和1.128±0.169,E-cadherin mRNA分别为1.125±0.092和0.352±0.036,Vimentin mRNA分别为1.253±0.130和2.875±0.366,差异均具有统计学意义(P<0.05)。两组MMP-2蛋白表达量分别为0.254±0.031和0.839±0.073,MMP-9蛋白表达量分别为0.268±0.039和0.733±0.075,MMP-2 mRNA分别为1.230±0.125和4.369±0.368,MMP-9 mRNA分别为1.452±0.023和4.136±0.569,差异均具有统计学意义(P<0.05)。结论 缺氧可通过调控转移相关基因的表达促进胰腺癌细胞的侵袭和迁移。
