The Diagnosis and Prognostic Significance of miR-326 in Childhood Acute Lymphoblastic Leukemia

儿童急性淋巴细胞白血病中miR-326的表达及临床意义

Abstract: Objective This study aims to investigate the diagnosis and prognostic significance of miR-326 in childhood acute lymphoblastic leukemia. Methods 70 blood samples from childhood ALL patients and 30 blood samples from healthy volunteers were collected, and qRT-PCR was adopted to detect the relative expression level of serum miR-326. The ROC curve was used to analyze the diagnostic ability and prognostic value of miR-326 in childhood ALL. At the same time, Kaplan-Meier was also performed to draw survival curves to evaluate the survival of children with ALL patients with different expression levels of miR-326. Results Compared with the control group, the expression of miR-326 in the serum of childhood ALL patients was significantly down-regulated (P<0.05). The ROC curve showed an AUC value of 0.876 for the area under the curve, indicating that miR-326 has a good diagnostic value; overall survival analysis showed that low expression of miR-326 suggested a poor survival rate, and the ROC curve showed an AUC value of 0.909. Analysis of progression-free survival showed that low expression of miR-326 suggested a poor progression-free survival rate, and the ROC curve showed an AUC value of 0.815. This indicated that low expression of miR-326 is associated with a poor prognosis. Conclusion According to the expression of miR-326, it is obvious to distinguish children with ALL from healthy people. In addition, the low expression of miR-326 indicates a poor prognosis in children with ALL. Furthermore, miR-326 can be used as a diagnostic and prognostic marker for childhood ALL.

摘要: 目的 本研究旨在探讨miR-326在儿童急性淋巴细胞白血病中的诊断及预后意义。方法 收集70份儿童ALL患者血液样本和30份健康志愿者的血液样本,采用qRT-PCR检测血清miR-326的相对表达水平。采用ROC曲线分析miR-326在儿童ALL中的诊断能力和预后价值,并采用Kaplan-Meier绘制生存曲线,评估miR-326表达水平不同的儿童ALL患者的生存情况。结果 与对照组相比,miR-326在儿童ALL患者血清中的表达明显下调(P<0.05)。ROC曲线显示曲线下面积AUC值为0.876,表明miR-326有较好的诊断价值;总生存分析显示,低表达的miR-326提示较差的生存率,ROC曲线显示AUC值为0.909。无进展生存分析显示,低表达的miR-326提示较差的无进展生存率,ROC曲线显示AUC值为0.815。表明miR-326低表达与较差的预后相关。结论 根据miR-326的表达情况可以很明显的将儿童ALL患者与健康人群进行区分。此外,miR-326低表达提示儿童ALL患者预后不良,即miR-326可作为儿童ALL的诊断及预后标志物。
