Expression of Serum ADPN, FKN and Gremlin in Diabetic Nephropathy Patients and Correlation with Renal Function

Objective To investigate the expression of serum adiponectin (ADPN), irregular chemokine (FKN) and Gremlin in diabetic nephropathy (DN) patients and their correlation with renal function. Methods One hundred cases of DN patients admitted to First Affiliated Hospital of Henan University of Science and Technology from June 2020 to June 2022 were retrospectively selected as the observation group, and another 100 cases of simple diabetes mellitus (DM) patients in the same period were selected as the control group. The serum levels of ADPN, FKN and Gremlin were compared between the two groups, and the renal function indexes [cystatin C (CysC), creatinine (SCr) and urinary protein excretion rate (UAER)] were compared between the two groups. The correlation between serum indexes and renal function indexes was analyzed, and the diagnostic value of serum indexes in DN was analyzed by ROC. Results Serum levels of ADPN, FKN, Gremlin, CysC, SCr and UAER in observation group were higher than those in control group (P < 0.05). Serum levels of ADPN, FKN and Gremlin were positively correlated with renal function indexes CysC, SCr and UAER, and the AUC of all serum indexes in combined diagnosis of DN was 0.885, the best diagnostic sensitivity and specificity were 90.00% and 85.00%, respectively, and the Yoden index was 0.750. Conclusion ADPN, FKN and Gremlin are up-regulated in the serum of patients with DN, and the levels of each indicator are positively correlated with the levels of renal function indicators. Combined detection has certain diagnostic value for DN, and can be used as an effective indicator for clinical diagnosis of DN and evaluation of renal function.

Influence of Percutaneous Vertebroplasty in the Treatment of Thoracolumbar Compression Fractures

Objective To investigate the effects of percutaneous vertebroplasty (PVP) on lumbar function, serum levels of prostaglandin E2 (PGE2) and substance P (SP) in patients with thoracolumbar compression fractures (TCF). Methods A total of 92 TCF patients who were treated in Xuchang Hospital of Traditional Chinese Medicine from January 2021 to May 2022, were collected and randomly divided into two groups, 46 cases in each group. The control group underwent percutaneous kyphoplasty (PKP), and the observation group underwent PVP. Perioperative indexes, serum levels of PGE2 and SP , imaging parameters (vertebral anterior margin height, vertebral Cobb angle), Oswestry disability index (ODI) before and after operation, and complications were compared between the two groups. Results The operation time, fluoroscopy times and hospitalization days in the observation group were less than those in the control group, the differences were statistically significant (P<0.05). After 7 days of operation, serum levels of SP and PGE2 in two groups were decreased compared with before operation, and the differences were statistically significant (P<0.05). After operation, the vertebral anterior margin height and vertebral Cobb angle in the observation group were better than those in the control group, and the differences were statistically significant (P<0.05). After operation, there was no significant difference in ODI between the two groups (P>0.05). There was no significant difference in the total incidence of complications between the two groups (10.87% vs 21.74%) (P>0.05). Conclusion PVP therapy in treatment of TCF patients can reduce serum levels of PGE2 and SP, improve lumbar function and promote postoperative recovery.

Relationship between Serum Gal-3, SAA, IL-33 and the Condition of Children with Allergic Asthma

Objective To analyze the correlation between serum levels of galectin-13 (Gal-3), serum amyloid A protein(SAA), interleukin-33 (IL-33) and the severity of allergic asthma in children. Methods A total of 102 children with allergic asthma in Xuchang People's Hospital from September 2021 to June 2023 were selected as the study group, and 102 healthy children in the same period and at the same age were selected as the control group. The serum levels of Gal-3, SAA, IL-33, pulmonary function [forced vital capacity (FVC), peak expiratory flow (PEF)], and exhaled nitric oxide (FeNO) were compared between the two groups. The correlations of serum Gal-3, SAA, IL-33 levels with pulmonary function, FeNO, and disease severity were analyzed. Results The levels of serum Gal-3, SAA and IL-33 in the study group were higher than those in the control group (P < 0.05). PEF and FVC in the study group were lower than those in the control group, and FeNO was higher than that in the control group (P < 0.05). The serum levels of Gal-3, SAA, and IL-33 in mild children were lower than those in moderate and severe children (P < 0.05). PEF and FVC in the mild group > moderate > severe, FeNO mild < moderate < severe (P < 0.05). Serum levels of Gal-3, SAA and IL-33 were negatively correlated with PEF and FVC, and positively correlated with FeNO and disease severity (P < 0.05). Conclusion The levels of serum Gal-3, SAA and IL-33 in children with allergic asthma are up-regulated, which are closely related to lung function, airway inflammation and disease severity, and have important clinical significance in judging the severity of the disease.

Effects of Electroacupuncture on the Expression of Nogo-A/NgR in the Lateral Geniculate Nody of Rats with Monocular Deprivation Amblyopia

Objective To investigate the effect of electroacupuncture on the expression of Nogo-A and NgR proteins in the lateral geniculate body of monocular deprivation amblyopia rats. Methods Twenty-eight out of 40 SPF-grade newborn SD rats were randomly selected to establish a monocular deprivation rat model by right eyelid suture at 14d of birth, and 24 rats with successful modelling were randomly divided into the model group and the electroacupuncture group. In the electroacupuncture group, electroacupuncture was applied to the left and right sides of the eye at "Cuanzhu" and "Fengchi". Differences in the amplitude and latency values of the P100 wave of the P-VEP of the rats in each group were detected by P-VEP; HE staining was used to observe the pathological morphological changes of neurons in LGN; immunohistochemistry and Western blot were used to detect the expression levels of Nogo-A and NgR proteins in the lateral geniculate body. Results Compared with the normal group, the cells of the model group were solidified; the P100 wave amplitude in the deprived eye was significantly decreased and the latency value was significantly increased, the mean cell number of Nogo-A and NgR proteins in LGN was increased, the average gray value was decreased, and the relative expression was increased (P<0.01). In the electroacupuncture group, compared with the model group, the cellular of the deprived eye morphology had a significantly improved; the P100 wave amplitude was significantly higher (P<0.05), and the latency value was significantly reduced (P<0.01); and the mean number of positive cells in LGN for Nogo-A and NgR proteins was reduced, with an increase in the mean gray value (P<0.05) and a decrease in the relative expression (P<0.01). Conclusion Electroacupuncture protects nerve cells damaged by monocular deprivation of the posterior LGN, and its mechanism of action may be associated with inhibition of the over expression of Nogo-A and NgR.

Effect of Urantide on α-SMA and OPN in ApoE -/- Mice with Atherosclerosis combined with Metabolically Related Fatty Liver Disease

Objective To observe the effect of Urantide on α-SMA and OPN in ApoE -/- mice with metabolic-associated fatty liver disease (MAFLD), and to explore its therapeutic effect on liver fibrosis in atherosclerosis (AS) complicated with MAFLD. Methods ApoE -/- mice were divided into AS model group, simvastatin group, low -dose, medium -dose, and high-dose Urantide groups. C57BL/6 mice were used as control group. The control group and AS model group were treated with normal saline 20 mg/kg, the simvastatin group was treated with simvastatin 5 mg/kg, and the Urantide group was treated with Urantide 20 mg/kg, 30 mg/kg and 40 mg/kg, respectively. After 14 days of treatment, the pathological changes of liver were observed by HE and oil red O staining. Masson trichrome staining was used to observe the expression of collagen fibers in the liver. The expression and localization of α-SMA in liver were detected by immunohistochemistry. RT-qPCR was used to detect the gene changes of UⅡ/UT system. The expressions of α-SMA, OPN and UⅡ/UT system in liver were detected by Western blotting. Results Compared with the control group, the mice in the AS model group showed liver steatosis, oil red O staining showed a large number of lipid droplets infiltration, collagen fiber deposition in the periosinusitis space, and the expression of α-SMA, OPN, UII and GPR14 protein in the liver increased. UII and GPR14 mRNA levels were increased. After Urantide treatment, the lipid droplets and fatty lesions in the liver were significantly reduced, collagen fibers were reduced, and the expressions of UⅡ/UT system, α-SMA and OPN were significantly decreased. Conclusion Urantide may improve hepatic steatosis in ApoE -/- mice by antagonizing the UⅡ/UT system.
