Effect of Ganoderma Lucidum Polysaccharide on Interleukin-10 Production of B16F10 Melanoma Cells

Objective To study the effect of production of Interleukin-10(IL-10) in the B16F10 cells by Ganoderma lucidum polysaccharides.
Methods The B16F10 cell was prepared and treated with different concentrations of Ganoderma lucidum. The expression levels of IL-10 were assessed by Western-blot, RT-PCR.
Results Under Ganoderma lucidum polysaccharides at different concentrations, RT-PCR revealed that the amount of IL-10 mRNA released by B16F10 melanoma cells treated with 0μg/mL, 0.2μg/mL, 0.8μg/mL, 3.2μg/mL, 12.8μg/mL was 1.03±0.15, 1.15±0.25, 0.92±0.11, 0.86±0.64 and 0.75±0.12, respectively. Compared with the 0μg/mL Ganoderma lucidum polysaccharide group, the other groups showed statistical significance except the 0.2μg/mL Ganoderma lucidum polysaccharide group, P<0.05. Western-blot revealed that the amount of IL-10 produced by B16F10 melanoma cell treated with 0μg/mL,0.2 μg/mL, 0.8μg/mL, 3.2μg/mL, 12.8μg/mL was 1.47±0.43, 1.58±0.31, 0.93±0.12, 0.89±0.26 and 0.40±0.29, respectively. Compared with the 0μg/mL Ganoderma lucidum polysaccharide group, the 12.8μg/mL Ganoderma lucidum polysaccharide group showed statistical significance, P<0.05.
Conclusion The production of IL-10 in the B16F10 cells was inhibited by Ganoderma lucidum polysaccharides.
