SWE Combined with CT in the Evaluation of Muscle Quality of Chronic Ankle Instability

Objective
To evaluate the effect of shear wave elastography (SWE) combined with CT in the evaluation of chronic ankle instability (CAI).
Methods
Retrospective analysis was made on 50 patients with unilateral chronic ankle instability admitted to Chengde Central Hospital from October 2020 to October 2021. The patients were divided into healthy side group and affected side group according to their healthy side and affected side. CT and SWE were adapted to examine the medial head of gastrocnemius muscle and soleus muscle on the healthy side and the affected side of CAI patients. CT values (Hu), Young's modulus (Kpa) and other parameters were measured.
Results
There was a significant positive correlation between SWE and CT values in the evaluation results of chronic ankle instability muscles (pearson correlation coefficient of gastrocnemius muscle=0.661, P<0.001; pearson correlation coefficient of soleus muscle=0.726, P<0.001). Young's modulus measured by SWE gradually decreased, and CT value also gradually decreased, the difference was statistically significant (P<0.001).
Conclusion
SWE technique and CT examination are correlated in the evaluation of chronic ankle instability muscle, and the changes are consistent.

EFFECTS OF OSU-03012 ON PROLIFERATION AND INVASION OF HUMAN COLON CANCER LOVO CELLS AND THE EXPRESSION OF COX-2, VEGF, MMP-2

Objective:
To explore the effects of OSU-03012 on proliferation and invasion of human colon cancer LoVo cells and the expression of cyclooxygenase-2 (COX-2), vascular endothelial growth factor (VEGF) and matrix metalloproteinase-2 (MMP-2).
Methods:
The human colon cancer LoVo cells were treated with 0μmol/L, 3μmol/L, 5μmol/L and 10μmol/L of OSU-03012. The proliferation ability of LoVo cells in each group was detected by MTT assay, the invasion ability was detected by Transwell method. The expression of COX-2, VEGF and MMP-2 in LoVo cells were detected by qRT-PCR and Western Blot.
Results:
The colon cancer LoVo cells were treated with OSU-03012 for 24h, 48h and 72h, the proliferation ability of LoVo cells gradually decreased with the increasing of drug concentration; and pairwise comparison of different concentrations at the same time point showed statistically significant differences (P<0.05). After the LoVo cells were treated with OSU-03012 for 24h of different concentrations, the number of LoVo cells passing through the basilar membrane, as well as COX-2, VEGF, MMP-2 mRNA and protein expression decreased significantly; Pairwise comparison of different concentrations showed statistically significant differences (P<0.05).
Conclusions:
OSU-03012 can inhibit the proliferation and invasion of human colon cancer LoVo cells, and the molecular mechanisms may be related to down-regulation of COX-2,VEGF and MMP-2 expression.

Objective:
To explore the application value of superb microvascular imaging (SMI) in detecting the synovial blood vessels of knee joint in patients with rheumatoid arthritis (RA), by comparing with contrast-enhanced ultrasound (CEUS).
Methods:
SMI and CEUS were respectively used to detect and record the blood flow signals of synovium of knee joint of 52 RA patients undergoing total knee arthroplasty (TKA). The consistency of grading of synovial vessels by SMI and CEUS, as well as the correlation between SMI grading and pathological grading were analyzed.
Results:
There were 104 knee joints in 52 patients, 63 of them underwent total knee arthroplasty. SMI and CEUS all displayed 92.31% (96/104) of the synovial blood flow signals of the knee joint of RA patients, and the consistency between SMI and CEUS was high (Kappa=0.724). Spearman correlation analysis showed that SMI grading was positively correlated with pathological grading (r=0.744, P<0.05).
Conclusions:
Displaying the synovial blood flow signals of the knee joint of RA patients by SMI was consistent with CEUS; Moreover, SMI can provide some reference value for judging the activity of RA.
