Effect of hsa-circ-0001862 on the Phenotype of Tongue Squamous Cell

Objective To investigate the effect of circular RNA hsa-circ-0001862 on the phenotype of tongue squamous cell carcinoma and its mechanism on the occurrence and development of tongue squamous cell carcinoma. Methods hsa-circ-0001862 plasmid was constructed, and the interaction relationship between hsa-circ-0001862 and miR-23a-3p was verified by double luciferase reporter gene and qRT-PCR. After targeted inhibition of hsa-circ-0001862, CCK8 assay and colony formation assay were used to detect the cell viability and colony formation ability of tongue squamous cell carcinoma cells. The migration and invasion ability of tongue squamous cell carcinoma cells were detected by scratch test and Transwell test. Apoptosis-related proteins were detected by Western blot to reflect the effect of hsa-circ-0001862 on the apoptosis of tongue squamous cell carcinoma cells. Results hsa-circ-0001862 and miR-23a-3p could interact, and their expression was negatively correlated in tongue squamous cell, carcinoma cells. In Tca-8113 cells, hsa-circ-0001862 inhibited cell proliferation, migration and invasion (P<0.01), and promoted cell apoptosis (P<0.01). Conclusion hsa-circ-0001862 interacts with miR-23a-3p, and hsa-circ-0001862 plays an inhibitory role in the development of tongue squamous cell carcinoma. hsa-circ-0001862 may be a new biomarker and target for the treatment of tongue squamous cell carcinoma.

Predictive Value of Serum Betatrophin in Type 2 Diabetes Mellitus with Metabolic Related Fatty Liver Disease

Objective To observe the changes of Betatrophin levels in type 2 diabetes mellitus combined with metabolism-related fatty liver diseases, and explore its predictive value for the disease. Methods A total of 58 patients with T2DM were admitted to the Department of Endocrinology and Metabolism in The Second Affiliated Hospital of Bengbu Medical College from January 2021 to November 2021 were selected as the case group. According to whether they had combined metabolic related fatty liver disease, patients were divided into T2DM with MAFLD group and T2DM alone group. Healthy subjects were selected as control group. We compared the general data, serum Betatrophin, blood glucose and lipid profiles, and analyzed the association between Betatrophin and type 2 diabetes mellitus with metabolic related fatty liver disease. Results There were significant differences in BMI, ALT, FPG, HOMA-IR, HbA1c, TG, LDL-C, HDL-C, Betatrophin between case group and control group (P<0.05). There were significant differences in BMI, HOMA-IR, HbA1c, TG and Betatrophin between T2DM with MAFLD group and T2DM alone group (P<0.05). Correlation analysis found that Betatrophin was positively correlated with TG. Multivariate analysis identified Betatrophin with TG as an independent risk factor for combined metabolic related fatty liver disease in patients with type 2 diabetes (P<0.05). Conclusion Betatrophin levels are significantly increased in the serum of T2DM patients with MAFLD, which promotes disease initiation and progression, and has predictive value for MAFLD.

Correlation between CT Measurement of Left Atrial Diameter and Recurrence of Atrial Fibrillation after Radiofrequency Ablation

Objective To investigate the predictive value of CT measurements of left atrium and pulmonary vein diameter for recurrence after radiofrequency catheter ablation (RFCA) in patients with primary atrial fibrillation. Methods Sixty-six patients with paroxysmal atrial fibrillation who were diagnosed as atrial fibrillation in the Affiliated Hospital of Chengde Medical University and underwent radiofrequency ablation for the first time were selected as the lesion group, and 40 healthy people were selected as the control group. All patients underwent CT angiography of left atrium and pulmonary vein. The transverse diameter and area of left atrium, the diameter and area of pulmonary vein opening, and the ratio of atrium to vertebra were measured, and the data were analyzed between groups. CT examination was performed again 3, 6 and 12 months after operation. According to the follow-up of patients, the patients were divided into recurrence group and cure group. The data of recurrence group and cure group were compared and analyzed. Results There were significant differences in the area of right superior pulmonary vein (RSPV), the anterior and posterior diameter and area of right inferior pulmonary vein (RIPV), and the diameter and area of left superior pulmonary vein (LSPV) between the observation group and the control group (P<0.05). There were significant differences in the left atrio-vertebral area ratio (LAVA), the left atrio-vertebral diameter ratio (LAVD), the area of RSPV and the anterior and posterior diameter of RIPV between the recurrence group and the cure group (P<0.05). Logistic regression analysis showed that LAVA was an independent risk factor for RFCA recurrence. Conclusion CT measurements of left atrium and pulmonary vein are closely related to the recurrence rate of atrial fibrillation after RFCA, and LAVA can be used as an independent predictor of recurrence after RFCA.

The Significance of Dsg1 and Dsg3 Antibody's Detection in Pemphigus Patients

Objective In order to investigate the application of anti-Dsg1and anti-Dsg3 antibody in diagnosis and treatment of patients, the level and changes of anti-Dsg1and anti-Dsg3 antibody in 18 patients with pemphigus and the relationship between the antibody index and the severity of the disease were analyzed. Methods 18 patients were diagnosed for pemphigus by direct immunofluorescence and pathology, and the patient's pathogenetic condition was assessed by ABSIS standard.The level of Dsg1 and Dsg3 of patients and nomals were detected by enzyme-linked immunosorbent assay(ELISA). Statistics was performed to analyze the relationship and significance of each index at last. Results Among the 18 patients, 13 were Dsg1 positive and 10 Dsg3 positive . The index of Dsg1 and Dsg3 after treatment were decreased than before, and the difference was statistically significant (P<0.05). Of the 20 nomals, 2 were Dsg1 positive and 1 was Dsg3 positive. The sensitivity of Dsg1 and Dsg3 antibody detection was 72.22% and 55.56% respectively, and the specificity was 90% and 95% respectively. The index of Dsg antibody was correlated with the ABSIS score (ALL P<0.05). Conclusion The Dsg antibody level of patients with pemphigus was correlated with the severity of the disease with high sensitivity and specificity. The detection of Dsg antibody level can be used for diagnosis and guidance of state and treatment of the disease.

GROWTH-PROMOTING EFFECTS OF RECOMBINANT HUMAN GROWTH HORMONE ON CHILDREN WITH GHD AND ISS AND ITS EFFECTS ON BLOOD LIPIDS

Objective: To investigate the effects of recombinant human growth hormone (rhGH) on body height and blood lipid of children with growth hormone deficiency (GHD) and idiopathic short stature (ISS). Methods: 37 children with GHD and 35 children with ISS were all treated with rhGH subcutaneous injection regularly. All the children were followed up every 3 months for 12 months, body height and blood lipid were measured during follow-up. Results: After treatment, the body height of GHD children and ISS children were all significantly higher than that before treatment (P<0.05). After treatment, the total cholesterol and triglyceride of GHD children and ISS children after treatment had no significant differences compared with that before treatment (P>0.05). After treatment, the high-density lipoprotein of GHD children and ISS children were significantly higher than that before treatment, while the low-density lipoprotein were significantly lower (P<0.05). Conclusions: rhGH treatment can significantly improve the body height and blood lipid indexes of GHD and ISS children, so it is worthy of clinical application.

CHANGES AND SIGNIFICANCE OF CD4 AND CD8 EXPRESSION IN SKIN LESIONS OF PATIENTS WITH ERYTHEMA NODOSUM

Objective: To study the changes and significance of CD4 and CD8 expression in skin lesions of patients with erythema nodosum. Methods: MaxVision TM immunohistochemical stain was used to detect the expression of CD4 and CD8 in skin lesions of 49 cases of erythema nodosum patients (disease group) and 10 cases of normal skin (control group). Results: The CD4 positive rate of disease group was 100.00%, which was obviously higher than control group (P<0.01). There was no significant difference of positive rate of CD8 expression between disease group and control group (P>0.05). Conclusions: There is an imbalance of T cells subgroups in skin lesions of erythema nodosum patients, which suggests that the cellular immune response mediated by CD4 + T cells plays an important role in the pathogenesis of erythema nodosum.

INTERVENTION EFFECTS OF PARENTERAL NUTRITION WITH DIFFERENT NITROGEN-CONTENT IN PATIENTS WITH GASTROINTESTINAL TUMORS AFTER SURGERY

Objective: To evaluate the intervention effects of parenteral nutrition with different nitrogen-content onnutriture and immune indice of patients with gastrointestinal tumors after surgery. Methods: 50 patients with gastrointestinal tumors after surgery were randomly divided into experimental group and control group with 25 patients in each group. All the patients were given total parenteral nutrition through central vein on the first day after operation, and the non-protein calorie were all 83.7-104.6kJ/(kg•d). Meanwhile, the nitrogen-content of experimental group was 0.20g(0.15-0.22g)/(kg•d), while the control group was 0.10g(0.09-0.11 g)/(kg•d). The hemoglobin, albumin, CD 3 +, CD 4 + and CD 4 +/CD 8 + of patients in 2 groups on the 14 th day after operation were compared. Results: 14 th day after operation, the hemoglobin, albumin, CD 3 +, CD 4 + and CD 4 +/CD 8 + of patients in experimental group were all obviously higher than control group (P<0.05). Conclusions: On the basis of giving the same heat quantity, total parenteral nutrition provided by normal nitrogen-content [0.20g/(kg•d)] can improve the nutritional and immune status of patients after gastrointestinal tumor surgery.
