REGULATING EFFECTS OF INTRAVENTRICULAR INJECTION OF LEPTIN ON MMP-7 PROTEIN IN UTERINE FLUID OF OEP RATS

Objective: To observe the effects of intraventricular microinjection of leptin on matrix metalloproteinases-7 (MMP-7) in uterine fluid of OEP rats. Methods: OEP rats model was divided into leptin injection group and normal saline injection group with 8 rats in each group. 4μl leptin and 4μl normal saline were injected into the lateral ventricle of rats in the 2 groups respectively; and 6 hours after injection, the uterine fluid of rats were obtained. Two-dimensional gel electrophoresis and MALDI-TOF-MS were used to separate and identify the differential proteins in uterine fluid; At last, Western blot was used to compare the MMP-7 expression in uterine fluid of rats in leptin injection group and normal saline injection group to verify the results of MALDI-TOF-MS. Results: The differential target protein separating by two-dimensional gel electrophoresis was MMP-7 identified by MALDI-TOF-MS. Western blot showed that the MMP-7 expression in uterine fluid of rats in leptin injection group was obviously higher than normal saline injection group (P<0.05). Conclusions: Microinjection of leptin into lateral ventricle can positively regulate the MMP-7 protein expression in uterine fluid of OEP rats for a certain amount of time.

EFFECTS OF ACUPUNCTURE ON CALCIUM CHANNEL CAV1.3 EXPRESSION AND NEURONAL APOPTOSIS OF SPINAL NUCLEUS OF TRIGEMINAL NERVE OF CHRONIC CEREBRAL ISCHEMIA RATS

Objective: To investigate the effects of acupuncture on calcium channel Cav1.3 expression and neuronal apoptosis of spinal nucleus of trigeminal nerve of chronic cerebral ischemia rats. Methods: 36 male Wistar rats were divided randomly into acupuncture group, ischemia group and sham operation group. The chronic cerebral ischemia rats' model was made by occluding the left common carotid artery and bilateral vertebral arteries; and then the rats in acupuncture group were acupunctured the “Sibai acupoints” for 15 days. Immunohistochemical stain was used to detect the calcium channel Cav1.3 expression in spinal nucleus of trigeminal nerve, TUNEL to detect apoptosis of neurons, electron microscope to observe the ultrastructure of neurons. Results: The calcium channel Cav1.3 protein expression in spinal nucleus of trigeminal nerve of rats in acupuncture group was obviously higher than ischemia group, but still lower than sham operation group(P<0.05). The apoptotic neurons number of three groups from high to low was ischemia group, acupuncture group and sham operation group (P<0.05). The coated vesicles could be found in neurons of rats in acupuncture group under electron microscope. Conclusions: Acupuncture of rats' “Sibai acupoints” can obviously up regulate the calcium channel Cav1.3 expression and decrease neuronal apoptosis in spinal nucleus of trigeminal nerve of chronic cerebral ischemia rats.

REGULATING EFFECTS OF GNIH ON POMC IN HYPOTHALAMUS OF OEP RATS

Objective: To explore the effects of microinjection of gonadotropin releasing hormone (GnIH) into lateral ventricle on proopiomelanocortin (POMC) expression in hypothalamus of ovariectomized and estrogen-primed (OEP) rats.Methods:40 female SD rats were established OEP rat’s model and then randomly divided into control group and experimental group with 20 rats in each group. The rats in 2 groups were injected 4μl normal saline and 4μl GnIH into the lateral ventricle respectively; and the rats in each group were further divided into 1h, 2h, 4h, 6h 4 subgroups according to the time after injection (n=5). Western blot was used to detect the POMC protein expression in hypothalamus of rats in each group. Results: 1h, 2h, 4h after injection, there was no statistically significant difference about POMC expression in hypothalamus of rats in experimental group compared with that of rats in control group (P>0.05). 6h after injection, the POMC expression in hypothalamus of rats in experimental group was obviously lower than that of rats in control group (P<0.05). Conclusions: Microinjection of GnIH into the lateral ventricle can inhibit expression of POMC in hypothalamus.
