FEASIBILITY STUDY OF 80 KV COMBINED WITH SAFIRE TECHNOLOGY IN REDUCING CHEST CT RADIATION DOSE IN INFANTS

Objective: To study the feasibility of 80kV combined with original data-based iterative technology (SAFIRE) in reducing chest CT radiation dose in infants. Methods: 68 infants with chest CT scan were randomly divided into conventional dose group (n=34) and low dose group (n=34). The infants in conventional dose group underwent chest CT examination with 100kV tube voltage and filtered back projection (FBP) reconstruction; the infants in low dose group underwent chest CT examination with 80kV tube voltage and SAFIRE reconstruction. The image quality and radiation dose of 2 groups were compared. Results: There was no statistically significant difference in subjective image quality and objective image quality between conventional dose group and low dose group (P>0.05). The radiation dose of infants in low dose group was significantly lower than conventional dose group (P<0.05). Conclusions: 80kV tube voltage combined with SAFIRE technology can be used to reconstruct chest CT of infants, the image quality not only can be used for diagnosis, but also can significantly reduce the radiation dose, which can be regarded as the best chest CT of infants.

COMPARISON OF THE CURATIVE EFFECTS OF C 3-7 AND C 4-7 CERVICAL SINGLE DOOR LAMINOPLASTY IN THE TREATMENT OF CERVICAL SPONDYLOTIC MYELOPATHY

Objective: To evaluate the effects of cervical single-door laminoplasty that preserving insertion of semi-spinus muscle on C 2 vertebral body for treating cervical spondylotic melopathy. Methods: 60 patients with cervical spondylotic myelopathy were divided into group A and group B according to different surgical methods, 30 patients in each group. The patients in group A received C 3-7 cervical single-door laminoplasty, the patients in group B received C 4-7 cervical single-door laminoplasty. The JOA scores of patients before and at the last follow-up were analyzed, and the improvement rate of neurological function was calculated; The cervical curvature (Cobb angle) of the two groups before and at the last follow-up were compared; And the incidence rate of postoperative axial symptoms, C 5 nerve root paralysis were recorded. Results: At the last follow-up, the cervical curvature of patients in 2 groups was obviously lost compared with that before operation (P<0.05); At the last follow-up, the Cobb angle of patients in group A was obviously lower than group B (P<0.05). There was no significant difference in JOA score before and after operation, neurological improvement rate and incidence rate of complications between group A and group B (P>0.05). The JOA score of patients in 2 groups after operation were all obviously higher than than before operation (P>0.05). Conclusions: Treating cervical spondylotic myelopathy with C 3-7 and C 4-7 cervical single door laminoplasty all can get significant recovery of neurological function. While the cervical single-door laminoplasty with preservation of hemi-spinus muscles insertion on C 2 vertebral body not only can maintain the cervical curvature, but also can decrease the incidence rate of axial symptoms.

CHANGES OF IRS-1 IN THE LIVER OF TYPE 2 DIABETIC RATS AND THE REGULATORY ROLES OF SERICIN

Objective: To investigate changes of insulin receptor substrate-1 (IRS-1) in the liver of type 2 diabetic rats and the regulatory role of sericin. Methods: Thirty-six specific pathogen free (SPF) male rats were randomly divided into normal group (12 rats) and modeling group (24 rats). The rat’s model of type 2 diabetes mellitus was induced by high-fat, high-sugar feeding and intraperitoneal injection of streptozotocin. After modeling, the rats were randomly divided into model group (12 rats) and experimental group (12 rats). The rats in experimental group were given sericin for 35 days and other rats were given equal volume normal saline for 35 days. The glucose oxidase method was used to detect the fasting blood glucose of rats in each group. SP immunohistochemical staining and Real Time Q-PCR were used to detect the expression of IRS-1 in liver. Results: Compared with the rats in the model group, the blood glucose of rats in experimental group reduced significantly, and the expression of liver IRS-1 of rats in experimental group increased significantly (P<0.05). Conclusions: Sericin can enhance the effects of insulin signaling by upregulating the expression of IRS-1 in liver of diabetes mellitus rats, thus reducing the blood glucose.

EFFECTS OF TWO TREATMENTS ON VITRIFICATION OF MOUSE OVARIAN TISSUE

Objective: To compare the effects of fragment cryopreservation of mouse ovarian tissue and whole cryopreservation of mouse ovarian tissue on vitrification. Methods: Mouse ovarian tissues were randomly divided into fragment cryopreservation group, whole cryopreservation group and fresh control group. The ovarian tissue of mice in fresh control group were immediately fixed with 4% paraformaldehyde after taking out; The ovarian tissue of mice in fragment cryopreservation group were cryopreserved after cut into 1×1×1 mm 3 cubes; The ovarian tissue of mice in whole cryopreservation group were cryopreserved directly with no cutting. The rate of normal morphological follicles were calculated and the diameter of all kinds of follicles were measured after the ovarian tissues embedding and HE staining as normal. Results: The normal morphological follicles rate of ovarian tissues in fragment cryopreservation group and whole cryopreservation group were significantly lower than fresh control group (P<0.01); There had no statistical significance about normal morphological follicles rate between fragment cryopreservation group and whole cryopreservation group (P>0.05). The diameter of Primordial follicle, primary follicle, secondary follicle, sinus follicle of ovarian tissues in fragment cryopreservation group and whole cryopreservation group were significantly higher than fresh control group (P<0.05); There had no statistical significance about diameter of all kinds of follicles between fragment cryopreservation group and whole cryopreservation group (P>0.05). Conclusions: Whole cryopreservation of mouse ovarian tissue does not affect the effects of vitrification.

EFFECTS OF STILBENE GLYCOSIDES ON TTC29 GENE IN AGING LEYDIG CELLS

Objective: To investigate the effects of stilbene glycosides on Ttc29 gene in aging Leydig cells. Methods: The rats’ Leydig cells were isolated and primarily cultured. The model of Leydig cells senescence was established by free radical oxidative damage and identified by detecting the cell aging marker β-galactosidase. Methylation PCR and fluorescence quantitative PCR were used to detect the methylation status and transcription level of Ttc29 gene. Chromatin immunocoprecipitation was used to detect the enrichment of H3K27me3 and H3K9me3 in the promoter region of Ttc29 gene. Results: Compared with the cells in normal group, the positive rate of senescent cells in the aging group increased significantly (P<0.05); Stilbene glycosides could significantly reduce the positive rate of senescent cells (P<0.05). Compared with the normal group, Ttc29 gene methylation was significantly increased and mRNA expression level was significantly decreased in aging Leydig cells (P<0.05); And there had a negative correlation between them (P<0.05). Stilbene glycosides could significantly reduce the methylation of Ttc29 gene and increase mRNA expression level in aging leydig cells (P<0.05). No enrichment of H3K27me3 and H3K9me3 was detected in the promoter region of Ttc29 gene. Conclusions: Stilbene glycosides can inhibit the senescence of Leydig cells by changing the methylation level and mRNA expression level of Ttc29 gene.

PREDICTIVE VALUE OF THE CHANGES OF COAGULATION AND FIBRINOLYSIS FUNCTION IN PATIENTS WITH LUNG CANCER BEFORE AND AFTER RADIOTHERAPY FOR THE SHORT-TERM EFFICACY AND SURVIVAL PERIOD

Objective: To investigate the predictive value of the changes of coagulation and fibrinolysis function indexes in patients with lung cancer before and after radiotherapy for the short-term efficacy and survival period. Methods: 82 patients with lung cancer were all treated with IMRT, and the efficacy was evaluated after 3 months. The changes of D-dimer, fibrinogen (FIB), fibrin degradation products (FDP) and platelet (PLT) before and after radiotherapy were compared; And the relationships between the above indexes and patients' short-term efficacy, survival period were analyzed. Results: In 82 patients, 42 patients with partial remission (PR) after 3 months, 21 patients was stable (SD) and 19 patients was progression (PD). For PR patients (effective group), the D-dimer, FDP and PLT levle after radiotherapy were significantly lower than those before radiotherapy (P<0.05). For PD+SD patients (ineffective group), the D-dimer, FIB and FDP levle after radiotherapy were significantly higher than those before radiotherapy, but the PLT level was significantly lower (P<0.05). After radiotherapy, the D-dimer and FIB level of patients in effective group were significantly lower than ineffective group (P<0.05). The median survival period of 82 patients was (19.60±8.92) months. Before radiotherapy, the D-dimer, FIB, FDP of patients in good prognosis group (survival period>19 months) were significantly lower than poor prognosis group (survival period<19 months,P<0.05); There has no significant difference in PLT between good prognosis group and poor prognosis group (P>0.05). Conclusions: Monitoring the changes of D-dimer, FIB, FDP and PLT before and after radiotherapy in patients with lung cancer is helpful to evaluate the short-term efficacy and survival period.

EXPRESSION AND SIGNIFICANCE OF APOPTOSIS FACTOR FADD PROTEIN AND CASPASE-7 PROTEIN IN GASTRIC ADENOCARCINOMA

Objective: To investigate the expression and significance of Fas-associated death domain (FADD) protein and Caspase-7 protein in gastric adenocarcinoma Methods: The expression of FADD protein and Caspase-7 protein in 60 cases of gastric adenocarcinoma and 50 cases of normal gastric mucosa were detected by immunohistochemical SP method. The relationships between FADD protein, Caspase-7 protein and clinicopathological features of gastric adenocarcinoma, as well as the correlations between FADD protein and Caspase-7 protein in gastric adenocarcinoman were analyzed. Results: The positive products of FADD protein and Caspase-7 protein were all brownish yellow granules, mainly located in the cytoplasm. The positive expression rate of FADD protein and Caspase-7 protein in gastric adenocarcinoma were significantly lower than that in normal gastric mucosa tissues (38.3% vs 60.0%, 46.7% vs 68.0%, P<0.05). In gastric adenocarcinoma, the FADD protein and Caspase-7 protein expression were not correlated with age and gender (P>0.05); but related to the differentiation, TNM stage, lymph node metastasis and infiltration depth (P<0.05). Spearman correlation analysis showed that there had positive correlation between expression of FADD protein and caspase-7 protein in gastric adenocarcinoma (r=0.362, P<0.05). Conclusions: Low expression of FADD protein and Caspase-7 protein may play a synergistic role in the development of gastric adenocarcinoma.

INHIBITING EFFECTS OF PICROPODOPHYLLIN ON HUMAN BREAST CANCER MCF-7 CELL PROLIFERATION

Objective: To investigate the inhibiting effects of picropodophyllin on human breast cancer MCF-7 cell proliferation. Methods: Human breast cancer MCF-7 cells were cultured in vitro, and MTT method was used to detect the inhibitory effects of different concentrations of picropodophyllin (0.1μmol/L, 0.2μmol/L, 0.4μmol/L, 0.8μmol/L, 1.6μmol/L,3.2μmol/L, 6.4μmol/L) on proliferation of MCF-7 cells for 24h, 48h and 72h. At the same time, the structural changes of MCF-7 cells were observed under inverted microscope at different concentrations of picropodophyllin for 48h. Results: The concentration range was 0.1μmol/L~ 6.4μmol/L; and the growth inhibiting rate of MCF-7 cells increased gradually with the increasing of picropodophyllin concentration and action time (P<0.05). The IC 50 of MCF-7 cells treated with picropodophyllin for 24h, 48h and 72h were 7.32μmol/L, 0.720μmol/L and 0.318μmol/L respectively. Under inverted microscope, the MCF-7 cells contracted with unclear structure and the refractivity reduced after treated with picropodophyllin for 48h. Conclusions: Picropodophyllin has significant inhibiting effects on human breast cancer MCF-7 cells.

EXPERIMENTAL STUDY ON BONE MARROW MESENCHYMAL STEM CELL EXOSOMES REGULATING STAT3/GAP43 PATHWAY TO PROMOTE AXONAL GROWTH OF PRIMARY SENSORY NEURONS

Objective: To explore the effects of bone marrow mesenchymal stem cell (BMSC) exosomes on STAT3/GAP43 pathway and axonal growth of primary sensory neurons. Methods: BMSC of Wistar rats was extracted and the cell morphology was observed and identified by light microscope. Hypercentrifugation was used to extract BMSCs exosomes and identified the exosomes. Primary sensory neurons (rat dorsal root ganglion neurons) were treated with BMSC exosomes, immunofluorescence staining was used to detect axon growth of primary sensory neurons, and Western Blotting to detect STAT3 and GAP43 protein expression in primary sensory neurons. Results: Immunofluorescence staining showed that the extracted cells expressed CD29 and CD90 simultaneously, which proved the extracted cells were BMSC. Western Blotting showed the extracted exosomes expressed HSP70 and Alix. Compared with the control group, the length of neuronal axon in BMSC exosomes group increased significantly, the expression of STAT3 and GAP43 protein also significantly increased in BMSC exosomes group (P<0.05). Conclusions: BMSC exosomes can promote axonal growth of primary sensory neurons by regulating STAT3/GAP43 pathway.

CLINICAL EFFECTS OF IROOT BP ON REPAIRING FOR PERFORATION OF PULP FLOOR

Objective: To investigate the clinical effects of iRoot BP on repairing perforation of pulp floor. Methods: Forty patients with perforation of pulp floor were selected in this study and randomly divided into observation group (n=20) and control group(n=20). The patients in observation group were repaired with iRoot BP, while the patients in control group were repaired with mineral trioxide (MTA). The success ratio of the reparation of patients in 2 groups 3 months, 6 months, 12 months after treatment were compared, as well as the incidence of complications and satisfaction rate of repair treatment of 2 groups were recorded and compared. Results: The success ratio of the reparation of patients in observation group 3 months and 6months were significantly higher than control group (P<0.05); There was no significant difference about success ratio of the reparation 12 months after treatment of 2 groups (P>0.05). None of the patients in observation group suffered occlusal pain and loose teeth, and the complication rate of observation group was 0.00%, which was significantly lower than control group (20.00%, P<0.05). The satisfaction rate of repair treatment of observation group was 95.00%, which was significantly higher than control group (65.00%, P<0.05). Conclusions: The treatment effects of iRoot BP for treating perforation of pulp floor are better than MTA, the safety and satisfaction rate of repair treatment are all higher, so it is worthy of clinical application.

EXPRESSION AND CLINICAL SIGNIFICANCE OF MIR-15A-5P AND MDR1 MRNA IN COLORECTAL CANCER

Objective: To study the expression and clinical significance of miR-15a-5p and multidrug resistance gene 1(MDR1) mRNA in colorectal carcinoma tissues. Methods: RT-qPCR was used to detect the expression of miR-15a-5p and MDR1 mRNA in 44 cases of colorectal carcinoma and adjacent non-tumor tissues. The relationships between miR-15a-5p, MDR1 mRNA and clinical pathological features of colorectal carcinoma, as well as the correlations between miR-15a-5p and MDR1 mRNA in colorectal carcinoma were also analyzed. Results: Compared with the adjacent non-tumor tissues, miR-15a-5p mRNA expression was significantly lower in colorectal carcinoma, but MDR1 mRNA expression was significantly higher (P<0.05). The miR-15a-5p and MDR1 mRNA expression were all concerned with lymph node metastasis and TNM stage of colorectal carcinoma (P<0.05). In colorectal carcinoma, miR-15a-5p mRNA expression was negatively correlated with MDR1 mRNA expression (r=-0.343, P<0.05). Conclusions: The expression of miR-15a-5p was low and MDR1 was high in colorectal cancer tissues. Both of them may play an important role in development of colorectal carcinoma.

REGULATORY EFFECTS OF SERICIN ON BAX EXPRESSION IN RAT INS-1 CELL INDUCED BY STREPTOZOTOCIN

Objective: To observe the effects of sericin on Bax expression in rat insulinoma cell (INS-1 cell) injured by streptozotocin (STZ). Methods: INS-1 cells were divided into 3 groups: normal control group, the cells were not treated with any drugs; STZ treatment group, the cells were treated with STZ (10mmol/L) for 24h; sericin protection group, the cells were treated with STZ (10mmol/L) and sericin (300μg/ml) for 24h. CCK8 assay was used to measure the viability of INS-1 cells in each group; Western blot and real-time fluorescence quantitative PCR were used to detect the Bax protein and mRNA expression in INS-1 cells. Results: The viability of INS-1 cells in STZ treatment group was remarkably lower than normal control group, while the Bax protein and mRNA expression were remarkably higher (P<0.05). The viability of INS-1 cells in sericin treatment group was remarkably higher than STZ treatment group, while the Bax protein and mRNA expression were remarkably lower (P<0.05). Conclusions: Sericin has protective effects on INS-1 cells injured by STZ, the mechanisms maybe concerned with it can down regulate Bax expression.

CONTENT DETERMINATION OF OSTHOL IN OSTHOL-CHITOSAN NANOMICELLES

Objective: To establish the method to determine the osthol content in osthol-chitosan nanomicelles. Methods: The content of osthol in osthol-chitosan nanomicelles was determined by high performance liquid chromatography by using Agilent C 18 column (24.6×200mm, 5μm), methanol-0.5% formic acid (75:25, v/v) as mobile phase, detection wavelength 322 nm, flow rate of 1.0ml/min, and the column temperature was 25℃. Results: The linear range for osthol was in the range of 0.78~50.00μg/ml and the precision of the method was good. The recoveries of low, medium and high concentration osthol were 101.7%, 98.2% and 98.9% respectively, and the RSD was lower 2%. The osthol content in three batches osthol-chitosan nanomicelles were 154.0μg/ml, 167.2μg/ml and 175.1μg/ml. Conclusions: The method established in this study has the advantages of simple, rapid and accurate, and it can be used for determining the osthol content in osthol-chitosan nanomicelles.

COMPARATIVE ANALYSIS OF ELISA AND CLIA IN DETECTION OF HBSAG AND SYPHILIS ANTIBODIES

Objective: To analyze the results of enzyme-linked immunosorbent assay (ELISA) and chemiluminescence immunoassay (CLIA) in detection of hepatitis B surface antigen (HBsAg) and Treponema pallidum (TP) antibody. Methods: ELISA and CLIA were used to detect HBsAg and TP antibody of 3060 blood samples. HBsAg negative samples were then detected by nucleic acid test (NAT). All the mismatching results of HBsAg and anti-TP detected by ELISA and CLIA were confirmed in National Center for Clinical Laboratories (NCCL). Results: HBsAg tests results: the sensitivity of ELISA and CLIA were all 90.00%, the specificity was 99.87% and 100.00% respectively. 4 cases ELISA positive CLIA negative samples were confirmed negative at last, but 1 positive sample of NAT was confirmed reactive. TP antibody tests results: the sensitivity of ELISA and CLIA was 40.00% and 90.00% respectively, the specificity was 99.84% and 99.97% respectively. 12 ELISA reactive CLIA negative samples were confirmed negative in 11 cases and reactive in 1 case; 8 CLIA positive ELISA negative samples, 6 samples were confirmed reactive while the other 2 were negative. Conclusions: CLIA has high sensitivity and specificity in detecting HBsAg and TP antibody of blood samples.

CLINICAL ANALYSIS OF DIAGNOSING AND TOTAL THORACOSCOPIC SURGERY FOR MEDIASTINAL CYSTS OF 32 CASES

Objective: To explore the clinical features, diagnosis and thoracoscopic surgery key points of mediastinal cysts. Methods: The clinical data, diagnosis and thoracoscopic surgery key points of 32 mediastinal cysts patients that were treated with totally thoracoscopic surgery from 2011.1 to 2018.4 were retrospectively analyzed. Results: Preoperative CT showed that 27 cysts in anterior mediastinum, 1 in middle mediastinum, 4 in posterior mediastinum; and the longest diameter of the cysts was (29.83±17.59)mm. The plain scan CT value of cysts was (25.03±20.03)HU, the enhanced CT value was (33.10±18.56)HU.The CT value of cysts in plain and enhanced scans were not statistic different (t=-1.603, P=0.114). All the 32 patients were treated with thoracoscopic surgery, the average operative time was (104.33±20.51)min, the intraoperative blood loss was (20.15±17.20)ml, and no perioperative deaths and no patients of conversion to open surgery. The pathological diagnoses after operation were 25 cases thymic cyst, 2 cases pericardial cyst, 2 cases esophageal cyst, 1 case cystic mature teratoma, 1 case bronchial cyst and 1 case cystic schwannoma. The mean follow-up time of 32 patients was 3.2 years, and no deaths and recurrent cases. Conclusions: Thoracoscopic surgery is an effective method for diagnosing and treating of mediastinal cyst, and has the advantages of safety, reliability and minimal invasiveness.

CLINICAL STUDY ON BILATERAL CENTRAL LYMPH NODE DISSECTION FOR UNILATERAL DIFFERENTIATED THYROID CANCER

Objective: To investigate the rationality of bilateral central lymph node dissection for unilateral differentiated thyroid cancer. Methods: 80 cases of unilateral differentiated thyroid cancer patients were all treated with radical thyroidectomy combined with bilateral central lymph node dissection, and the central lymph node metastasis and postoperative complications of patients were analyzed. Results: Among 80 patients, ipsilateral central lymph node metastasis in 32 patients (32/80, 40.0%), contralateral central lymph node metastasis in 8 cases (8/80, 10.0%), simultaneous metastasis of bilateral central lymph nodes in 16 cases (16/80, 20.0%). The total contralateral lymph node metastasis rate was 30.0% (24/80), the total lymph node metastasis rate was 70.0% (56/80). 9 (9/80, 11.3%) patients suffered temporary hypocalcemia, 8 (8/80, 10.0%) patients suffered temporary recurrent laryngeal nerve injury after operation. All the 80 patients were followed up for 3 years without recurrence or metastasis. Conclusions: Ipsilateral central lymph node is the most easily metastatic site of unilateral differentiated thyroid cancer, but contralateral central lymph node also has possibility of metastasis and skip metastasis. Bilateral central lymph node dissection for unilateral differentiated thyroid cancer can remove potential metastatic lymph nodes effectively and determine metastasis of the tumour, which is benefical for reducing recurrence after operation and guiding postoperative staging.

PREVENTIVE EFFECTS OF POLYGALA ON DORSAL ROOT GANGLIA NEURONS INJURY IN RATS WITH DIABETIC PERIPHERAL NEUROPATHY

Objective: To investigate the preventive effects of Polygala on sciatic nerve-related neurons injury of diabetic peripheral neuropathy (DPN) rats. Methods: The Wistar rats were randomly divided into 3 groups: blank control group, DPN model group and Polygala prevention group (n=12). The rats in DPN model group and Polygala prevention group were made streptozotocin induced diabetes model. Then the rats in Polygala prevention group were lavaged with Polygala (2.7g/kg/d) for 6 weeks. The DPN rats' model was established with SNCV˂30m/s as the standard. The immunohistochemical staining was used to detect the number of Bcl-2 and Bax positive cells in sciatic nerve-related dorsal root ganglia; Western Blot to detect Bcl-2 and Bax expression in dorsal root ganglia and TUNEL to detect apoptotic index of neurons in dorsal root ganglia. Results: Compared with blank control group, the Bcl-2 immuno-positive cells and Bcl-2 expression in dorsal root ganglia of DPN rats decreased obviously (P˂0.01); while the Bax immuno-positive cells, Bax expression and apoptotic index in dorsal root ganglia increased obviously (P˂0.01). Compared with DPN model group, the Bcl-2 immuno-positive cells and Bcl-2 expression in dorsal root ganglia of rats in Polygala prevention group increased obviously (P˂0.01); while the Bax immuno-positive cells, Bax expression and apoptotic index in dorsal root ganglia of rats in Polygala prevention group decreased obviously (P˂0.01). Conclusions: Polygala can inhibit neurons apoptosis in dorsal root ganglia of DPN rats by upregulating Bcl-2 expression and downregulating Bax expression, so has preventive effects on sciatic nerve-related neurons injury.

EFFECTS OF IOPROMIDE ON ION CHANNELS IN VENTRICULAR MYOCYTES

Objective: To observe the effects of non-ionic hypoosmotic contrast agent, iopromide on ion channels in ventricular myocytes. Methods: The ventricular myocytes were isolated and whole cell patch-clamp technique was used to record the current density, steady-state activation curve and inactivation curve of sodium current (I Na), L-type calcium current (I Ca,L) and instantaneous outward potassium ion current (I to) before and after iopromide administration. Results: Compared with that before iopromide administration, the current density of I Na and I Ca,L decreased obviously, the current density of I to increased obviously after iopromide administration. While iopromide has no obviously effects on steady-state activation curve and inactivation curve. Conclusions: Iopromide may induce malignant ventricular arrhythmia by changing the duration of action potential of ventricular myocytes.

EFFECTS OF MONTELUKAST COMBINED BUDESONIDE FORMOTELLO ON FENO ANDPULMONARY FUNCTION OF CVA PATIENTS

Objective: To observe the effects of Montelukast combined Budesonide formotello on exhaled nitric oxide (FeNO) and pulmonary function of cough variant asthma (CVA) patients. Methods: 60 CVA patients were randomly divided into observation group and control group with 30 patients in each group. The patients in control group were treated with Budesonide formoterol inhalation; While the patients in observation group were treated with Budesonide formoterol inhalation combined Montelukast. The FeNO and pulmonary function of patients in both groups were compared after treating for 4 weeks. Results: After treatment, the FeNO of patients in 2 groups were all significantly decreased (P<0.05); And the FeNO of patients in observation group after treatment was significantly lower than control group (P<0.05). After treatment, the forced expiratory volume in one second (FEV1), forced vital capacity (FVC), and FEV1/FVC of both groups increased significantly (P<0.05); And, the FEV1, FVC and FEV1/FVC of patients in observation group after treatment was significantly lower than control group (P<0.05). Conclusions: Compared with Budesonide formotello alone, treating CVA with Montelukast Combined Budesonide formotello can inhibit respiratory inflammation and improve pulmonary function better, so it is worthy of clinical application.

Objective: To explore the best cut-off point of orthostatic plasma aldosterone/plasma renin concentration (ADRR) for screening primary aldosteronism (PA). Methods: 80 cases of patients with unknown causes of hypertension were divided into PA group (combined with PA, n=20) and primary hypertension group (hypertension group, not combined with PA, n=60). The body height, body weight, blood pressure, blood potassium, orthostatic plasma aldosterone concentration (PAC), plasma renin activity (PRA) and plasma direct renin concentration (PDC) were measured respectively of all the patients; And the PAC/PRA (ARR) and ADRR were calculated. The ROC curve was drawn to determine the best cut-off point of orthostatic ADRR screening for PA. Results: The serum potassium level, PDC and PRA of patients in PA group were significantly lower than hypertension group, while the PAC, ADRR and ARR were significantly higher than hypertension group (P<0.05). The area under ROC curve of ARR was 0.698, and the diagnostic cut-off for screening PA was 63.77 [(pg/ml)/(μg/L/h)]. The area under ROC curve of ADRR was 0.882, and the diagnostic cut-off for screening PA was 2.55 (pg/ml)/(pg/ml). Conclusions: The diagnostic value of ADRR screening for PA is higher than that of ARR. It is suggested that ADRR can be used in clinical screening for PA.

EFFECTS OF ERLOTINIB ON CURATIVE EFFECTS AND SURVIVAL RATE OF NSCLC PATIENTS WITH DIFFERENT MUTANTS OF EGFR GENE

Objective: To study the effects of epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI) Erlotinib on curative effects and survival rate of non-small cell lung cancer (NSCLC) patients with different mutants of EGFR gene. Methods: 72 cases of advanced NSCLC patients with EGFR positive mutations were divided into study group (n=33) and control group (n=39) according to mutations in exon 19 or 21 of EGFR gene. All the patients were treated with Erlotinib Hydrochloride Tablets orally till disease progression or death. The short-term clinical effective rate, time to progression (TTP) and one-year survival rate of patients in 2 groups were compared. Results: The short-term clinical effective rate, one-year survival rate and average TTP of patients in study group were all obviously higher than control group (P<0.05). Conclusions: The curative effects and survival rate of NSCLC patients with exon 19 mutation were better than NSCLC patients with exon 21 mutation when treating NSCLC with EGFR-TKI Erlotinib. Which suggests that detection of different mutants of EGFR gene may be helpful in formulating individualized therapeutic regimen and predicting the therapeutic effect of EGFR-TKI Erlotinib.

RELATIONSHIPS BETWEEN BONE TURNOVER MARKERS AND DIABETIC MICROVASCULAR COMPLICATIONS IN TYPE 2 DIABETES MELLITUS

Objective: To investigate the relationships between bone turnover markers and diabetic microvascular complications in type 2 diabetes mellitus. Methods: 146 type 2 diabetes mellitus patients were divided into experimental group (77 patients, complicated with diabetic nephropathy and/or diabetic retinopathy) and control group (69 patients, no diabetic microvascular complications). The general informations of patients in 2 groups were recorded; and the blood biochemical indicators and bone turnover markers of patients were detected. Results: The course of disease of patients in experimental group was remarkably longer than control group (P<0.05); the carboxy-terminal collagen crosslinks (CTX) of patients in experimental group was remarkably higher than control group, the osteocalcin (OC) was remarkably lower (P<0.05). Logistic regression analysis showed that CTX and course of disease were risk factors of type 2 diabetes mellitus with microangiopathy, OC was protective factor. Conclusions: Bone turnover markers CTX and OC are associated with the risk of microvascular complications in patients with type 2 diabetes mellitus, which are expected to be markers for monitoring microangiopathy and bone metabolism in patients with type 2 diabetes mellitus.

EFFECTS OF TROPOMODULIN1 ON RENAL URINE CONCENTRATION IN MICE

Objective: To analyze the effects of tropomodulin1 (Tmod1) on urine concentration in kidney of mice. Methods: Tubule-specific Tmod1 knockout mice, Tmod1 flox/flox/Ksp-cre + (TFK) and its control mice Tmod1 flox/flox/Ksp-cre -(TF)were tanken as research object in this study. The urine of mice was collected and the 24-hour urine volume as well as urine osmolality of all the mice were measured at basic state, water deprivation and water loading using metabolic cage system. Results: At basic state, water deprivation and water loading, the 24-hour urine volume of TFK mice was remarkably lower than TF mice, while the urine osmolality of TFK mice was remarkably higher than TF mice (P<0.05). Conclusions: Tmod1 may be involved in the process of urine concentration in mice, but the mechanisms need to be further studied.

CHANGES OF NF-ΚB P65 EXPRESSION AND THE CORRELATIONS WITH INFLAMMATION IN SEBORRHEIC KERATOSIS

Objective: To investigate the changes of NF-κB P65 expression and its correlations with inflammation in seborrheic keratosis. Methods: Max VisionTM immunohistochemical stain and HE stain were used to detect the expression of NF-κB P65 and inflammation in 77 cases of seborrheic keratosis lesions, perilesional tissues and 10 normal skin tissues. Results: The positive expression rate of NF-κB P65 and the percentage of inflammation were 88.31% (68/77) and 80.52% (62/77) respectively, which were significantly higher than those of perilesional tissues and normal skin tissues (P<0.05). Spearman correlation analysis showed that NF-κB P65 expression is positively correlated to inflammation in seborrheic keratosis (r=0.769, P<0.05). Conclusions: High expression of NF-κB P65 and inflammation may play a synergistic role in development of seborrheic keratosis.

EFFECTS OF HESHOUWUYIN ON P53, BCL-2 AND BAX MRNA EXPRESSION IN AGING SPERMATOGENIC CELLS

Objective: To investigate the effects of Heshouwuyin on p53, Bcl-2 and Bax mRNA expression in aging spermatogenic cells. Methods: The spermatogenic cells aging model was established by free radical oxidative damaging method, and RT-qPCR was used to detect the p53, Bcl-2 and Bax mRNA expression in spermatogenic cells in each group. Results: The p53, Bax mRNA expression and Bax/Bcl-2 of spermatogenic cells in aging group were significantly higher than those in normal group (P<0.05),while Bcl-2 mRNA expression was significantly lower (P<0.05). The p53, Bax mRNA expression and Bax/Bcl-2 of spermatogenic cells in Heshouwuyin group were significantly lower than those in aging group (P<0.05),while Bcl-2 mRNA expression was significantly higher (P<0.05). Conclusions: Heshouwuyin can inhibit apoptosis of spermatogenic cells by upregulating Bcl-2 expression and downregulating p53 and Bax expression, so it can delay aging of spermatogenic cells.

Objective: To establish human choriocarcinoma JAR cells xenograft model in nude mice and observe its biological characteristics. Methods: 12 female Balb/c nude mice were randomly divided into normal control group and model group. The xenograft model was established by subcutaneous injection of choriocarcinoma JAR cells suspension; While the mice in normal control group were injected saline subcuatneouslly at the same site. The tumor formation rate and the survival time of nude mice were recorded; HE staining was used to observe the pathological changes of transplanted tumors and main organs, SP immunohistochemical staining to detect the β-human chorionic gonadotropin (β-HCG) expression in transplanted tumors of mice in model group, ELISA to detect the serum β-HCG level of mice in model group and normal control group. Results: The xenograft tumor formation rate in this study was 100% and the survival time of nude mice was (24.75±3.41) days. Gross morphological observation and histopathological observation showed that numerous trophoblastic proliferation with hemorrhagic necrosis in xenograft tumor, the cells markedly heteromorphic, with large and deeply stained nuclei. No obvious metastasis was observed in major organ. β-HCG protein positive expression could be observed in transplanted tumors of mice in model group. The serum β-HCG level of mice in model group was obviously higher than normal control group (P<0.05). Conclusions: The nude mice xenograft model is established using human choriocarcinoma JAR cells successfully in this study, which could provide an experimental animal model for choriocarcinoma studying.

Objective: To investigate the clinical effects of bedside ultrasound-guided central venous catheterization in patients with hypovolemic shock. Methods: 136 patients with hypovolemic shock that underwent central venous catheterization in ICU were divided into experimental group (bedside ultrasound-guided central venous catheterization) and control group (Seldinger puncture catheterization) according to catheterization mode. The catheterization time, success rate of puncture and incidence of complications of patients in 2 groups were compared. Results: The catheterization time of patients in experimental group was (7.2±1.8)min, which was obviously shorter than control group (10.6±2.1min, P<0.05). The first, second and third success rate of puncture of patients in experimental group were respectivley 94.3%, 98.6% and 100.0%, which were all obviously higher than control group (P<0.05). The incidence rate of local hematoma formation and mispuncture of artery of patients in experimental group were obviously lower than control group (P<0.05). Conclusions: Bedside ultrasound-guided central venous catheterization can increase the success rate of catheterization of patients with hypovolemic shock, also can shortern catheterization time and reduce complications, so has better application value.

ANAYLSIS OF THE RESULTS OF HPV INFECTION IN FEMALE GENITAL TRACT OF 13120 CASES IN CHENGDE

Objective: To analyze the genotype distribution and age distribution of human papillomavirus (HPV) infection of female in Hebei Chengde. Methods: Human papillomavirus nucleic acid typing detection kit was used to detect HPV infection of 13120 female in the Affiliated Hospital of Chengde Medical College, and the genotype distribution and age distribution of HPV infection were analyzed. Results: The HPV infection rate in 13120 female was 32.30% (4238/13120); 27 genotypes were detected, among which 16 genotype was the most, followed by 58, 52 and 6 genotype. In 4238 female, the constituent ratio of high-risk infection, low-risk infection and mixed infection were respectively 60.85%, 19.58% and 19.56%; the constituent ratio of single infection, double infection and multiple infection were respectivly 64.51%, 22.18% and 13.31%. The top three age groups of HPV infection rate were lower than 20 years old, 20-24 years old and 60-64 years old. Conclusions: The female HPV infection in Hebei Chengde area is mainly high-risk type and single infection, the genotypes with high infection rate were 16, 58, 52 and 6, and the distribution is U-shaped with age.

Objective: To explore the antihypertensive effects of taking antihypertensive drugs according to chronotherapeutics theory. Methods: 120 patients with primary hypertentsion were randomly divided into observation group and control group with 60 cases in each group. The patients in control group were given felodipine sustained-release tablets (5mg, qd) for l week, and then given candesartan (4mg, qd), both drugs were taken at 8:00 each day. The patients in observation group were also given felodipine sustained-release tablets and candesartan with the same dosage as cotrol group; but the two drugs were taken 3 hours before blood pressure peak according to 24-hour ambulatory blood pressure results. The therapeutic effects were evaluated using 24-hour ambulatory blood pressure results, including blood pressure changes, variation of blood pressure and circadian rhythm of blood pressure. Results: After treatment, the degree of 24-hour mean diastolic and systolic blood pressure, mean night systolic and diastolic blood pressure decline, proportion of blood pressure load below 10%, and proportion of Scoop type blood pressure of patients in observation group were all significantly higher than control group (P<0.05). Conclusions: The antihypertensive effects of taking antihypertensive drugs according to chronotherapeutics theory for treating primary hypertentsion patients were obviously better than traditional method; And it also can restore circadian rhythm of blood pressure and reduce blood pressure variation.

EFFECTS OF SERICIN ON PI3K MRNA EXPRESSION IN PANCREAS OF TYPE 2 DIABETES MELLITUS RATS

Objective: To investigate the effects of sericin on phosphatidylinositol-3-kinase (PI3K) mRNA expression in pancreas of type 2 diabetic rats. Methods: Male SD rats were randomly divided into three groups: normal control group, diabetic model group and sericin treatment group. The rat model of type 2 diabetes mellitus was established by feeding high-fat and high-sugar diet and intraperitoneal injection of streptozotocin (35mg/kg, twice). Then the rats in the sericin treatment group were given sericin (2.4g/kg/d) by gavage. Glucose oxidase method was used to detect the blood glucose of rats, and Real Time Q-PCR were used to detect the expression of PI3K mRNA in pancreas of rats in each group. Results: Compared with the normal control group, the blood glucose of diabetic rats increased significantly and the expression of PI3K mRNA in pancreas decreased significantly (P<0.05); Compared with diabetic rats, the blood glucose of sericin treated rats decreased significantly, and the expression of PI3K mRNA in pancreas increased significantly (P<0.05). Conclusions: Sericin may regulate insulin PI3K-Akt signaling pathway by increasing the expression of PI3K in pancreas, thus reducing blood glucose .
