Correlation Analysis of Serum ICAM-1, sLOX-1, and Lp-PLA2 Levels with Prognosis in Patients with Acute Cerebral Infarction

Objective To investigate the correlation between the expression levels of intercellular adhesion molecule-1 (ICAM-1), soluble lectin-like oxidized low-density lipoprotein receptor-1 (sLOX-1), and lipoprotein-associated phospholipase A2 (Lp-PLA2) in serum of patients with acute cerebral infarction (ACI) and their prognosis, and to analyze their predictive value for poor prognosis. Methods A retrospective study was conducted on 104 ACI patients who were diagnosed and treated in Zhengzhou Yihe Hospital from May 2021 to May 2023. After treatment, the prognosis was evaluated based on the modified Rankin Scale (mRS) at 90 days, and the patients were divided into a poor prognosis group (n=38) and a good prognosis group (n=66). The serum levels of ICAM-1, sLOX-1, and Lp-PLA2 were compared between the two groups. The influencing factors of prognosis were analyzed by multivariate Logistic regression. To evaluate the predictive value of serum ICAM-1, sLOX-1 and Lp-PLA2 in poor prognosis 1 month after treatment. Results The levels of serum ICAM-1, sLOX-1, and Lp-PLA2 in the poor prognosis group were higher than those in the good prognosis group 1 month after treatment (P<0.05). NIHSS score, neuron-specific enolase (NSE), central nervous specific protein (S100-β) and ICAM-1, sLOX-1 and Lp-PLA2 levels one month after treatment were independent risk factors for poor prognosis (P<0.05). The AUC of ICAM-1, sLOX-1 and Lp-PLA2 combined predicted poor prognosis at 1 month after treatment was greater than that of single index and any two indexes combined (P<0.05). Conclusion The levels of serum ICAM-1, sLOX-1, and Lp-PLA2 in ACI patients are independent risk factors for poor prognosis, and combined detection of their levels has certain predictive value for prognosis.

Diagnostic Value and Molecular Mechanism of LASP-1 in the Process of Advanced Adenoma and Carcinogenesis

Objective To detect the expression level of LASP-1 in advanced adenoma and adenocarcinoma and its diagnostic value, and to explore the molecular mechanism of malignant transformation of advanced adenoma. Methods Plasma samples were collected from 48 patients with colorectal adenocarcinoma (CRC group), 13 patients with adenoma (CRA group) and 22 healthy volunteers (Normal group). The expression level of LASP-1 in plasma samples was detected by enzyme-linked immunosorbent assay (ELISA). The expression of CEA in plasma was detected by automatic chemiluminescence immunoassay. The diagnostic efficacy of plasma LASP-1, CEA and their combination in CRC and CRA was evaluated by the area under the ROC curve. Spearman correlation analysis was used to analyze the relationship between plasma LASP-1 and clinicopathological features. The expression of LASP-1 in CRC stage I-IV patients and Normal group was detected in TCGA database. Next, the differential expression of LASP-1 in CRC and CRA groups was detected in GEO database. Immunohistochemical method and human protein immunohistochemistry database were used to detect the expression of P53 and LASP-1 in CRC tissues. Results ELISA results showed that the plasma concentrations of LASP-1 in CRC and CRA groups were significantly higher than those in Normal group (P<0.01), while there was no significant difference in CEA expression levels among CRC, CRA and Normal groups. ROC curve analysis showed that the sensitivity and specificity of plasma LASP-1 in the diagnosis of CRC and CRA were higher than those of CEA. When the two were combined, the area under the curve (AUC) was 0.982 and 0.99, respectively. TCGA database results showed that the expression level of LASP-1 in the tissues of CRC patients in each stage was significantly higher than that in the Normal group (P<0.05). GEO database results showed that the expression level of LASP-1 in the CRC group was significantly higher than that in the CRA group (P<0.05). Immunohistochemical results showed that mutant P53 accounted for 64.5% of the CRC group. Human protein immunohistochemistry database showed that LASP-1 was highly expressed in CRC tissues compared with normal intestinal epithelial tissues. Conclusion LASP-1 combined with CEA is a potential diagnostic indicator for early diagnosis of CRC and CRA patients; LASP-1 may be involved in the molecular mechanism of adenomasis cancer, making it possible for CRC to be detected and intervened in the adenoma stage.

Application of miR-493-5p, CHE combined with GP73 Detection in Screening of Liver Cancer and Disease Assessment

Objective To explore the application value of miR-493-5p, cholinesterase (CHE) combined with Golgiglycoprotein73 (GP73) detection in screening of liver cancer and disease assessment. Methods Ninety-eight patients with liver cancer (clinical stages: 31 cases in early stage, 35 cases in middle stage, and 32 cases in late stage) admitted to Anyang Fifth People's Hospital from July 2015 to July 2019 were selected as the observation group, and 104 patients who had healthy physical examination in this hospital during the same period were selected as the control group. The polymerase chain reaction-restriction fragment length polymorphism method was used to detect miR-493-5, and the butyrylthio-choline method was used to determine the level of CHE, and the electrochemiluminescence method was used to determine the level of GP73. The expression levels of the three indicators in different groups were compared, and the predictive value of the three combined detection on the progression of patients with liver cancer were analyzed. Results The levels of miR-493-5p and CHE in the observation group were lower than those in the control group, and the level of GP73 was higher than that in the control group, the differences were statistically significant (P<0.05). The relative expression of miR-493-5p and the level of CHE were compared: early stage>mid stage>mid-late stage, the differences were statistically significant (P<0.05). The level of GP73 was compared: early stage<mid stage<late stage, the difference was statistically significant (P<0.05). The sensitivity, specificity and AUC of the combined detection of miR-493-5p, CHE, and GP73 were 92.10%, 94.50%, and 0.960, respectively, which were higher than those of single index detection, and the differences were statistically significant (P<0.05). Conclusion In clinic, the combined detection of miR-493-5p, CHE, and GP73 can be strengthened for the diagnosis of early liver cancer and the disease evaluation of patients.

EFFECTS OF CORE STABILITY TRAINING COMBINED WITH BOBATH THERAPY ON LOWER LIMB FUNCTION AND BALANCE ABILITY OF STROKE PATIENTS

Objective: To investigate the effects of core stability training combined with Bobath therapy on lower limb function and balance ability of stroke patients. Methods: 44 stroke patients were randomly divided into control group and observation group with 22 patients in each group. The patients in control group were given conventional rehabilitation training; While the patients in observation group were also given core stability training combined with Bobath therapy. FMA scale, Sheikh scale, BBS scale, Holden walking function grading were used to evaluate the lower limb motor function, trunk control ability, balance ability and walking function of patients before training and 8 weeks training; Tape to measure the simple intra-abdominal pressure. Results: The FMA score, Sheikh score, BBS score and Holden score of patients in 2 groups after training were obviously higher than that before training (P<0.05); the intra-abdominal pressure was obviously lower (P<0.05). Moreover, the four scores and intra-abdominal pressure of patients in observation group improved significantly better than control group (P<0.05). Conclusions: Core stability training combined with Bobath therapy can obviously improve the lower limb function and balance ability of stroke patients on the basis of conventional rehabilitation training.
