Clinical Significance of P-selectin mRNA and Protein in the Differential Diagnosis of Benign and Malignant Hydatidform Mole

Objective To investigate the expression characteristics and clinical significance of P-selectin in benign and malignant hydatidiform mole tissues. Methods 50 cases of benign moles and 22 cases of malignant moles were selected. qRT-PCR technology is used to detect the expression level of P-selectin mRNA. Immunohistochemistry SP method and Western Blot technology comprehensively detected the expression level of P-Selectin protein. Results Compared with benign hydatidiform mole tissue, the relative expression of mRNA and protein and protein positive expression rate of P-selectin in malignant hydatidiform mole tissue were significantly higher (P<0.05). Conclusion P-selectin participates in the development of malignant hydatidiform mole at the gene and protein level, and is expected to become a diagnostic indicator for evaluating benign and malignant hydatidiform moles, providing new ideas for clinical targeted therapy.

Expression and Significance of Apoptosis Related Factor Bid Protein and mRNA in Gastric Adenocarcinoma

Objective To check the differential expression of apoptosis-related factor Bid in adenocarcinoma of stomach tissue an and analyze its clinical significance. Methods 60 cases of adenocarcinoma of stomach tissues were the experimental group, and 55 cases of normal gastric mucosal epithelium tissue were the control group. The expression of Bid protein and mRNA was comprehensively detected by combined immunohistochemical SP method, Western blot and qRT-PCR technology. Results Compared with normal gastric mucosal epithelium tissue, the positive expression level of Bid protein and the relative expression level of mRNA in adenocarcinoma of stomach tissue are reduced, and the differences are statistically significant (P<0.05). In protein level and gene level, the Bid molecule expression level in adenocarcinoma of stomach tissues was related to the degree of differentiation, lymph node metastasis, clinical TNM stage and infiltration depth(P<0.05). Conclusion The apoptosis-related factor Bid plays a vital role in the occurrence and development of adenocarcinoma of stomach, and it may become a new target for the treatment of adenocarcinoma of stomach.

Expression and Correlation of Caspase-9 and c-Iap1 in Gastric Adenocarcinoma

Objective By examining the protein expression of Caspase-9 and c-IAP1 in gastric adenocarcinoma tissues, to explore their relationship with the development and progression of gastric adenocarcinoma; and the correlation between the two. Methods The protein expressions of Caspase-9 and c-IAP1 were detected in 50 cases of gastric adenocarcinoma tissues and 40 cases of normal gastric mucosa tissues,byimmunohistochemical method. Results The expression of Caspase-9 protein in gastric adenocarcinoma was significantly lower than that of normal gastric mucosa(P<0.05);The expression of Caspase-9 protein in gastric adenocarcinoma tissues were significantly correlated with histological differentiation,TNM clinical stage and lymph node metastasis(P<0.05),while there was no correlation with gender, age and depth of invasion (P>0.05).The expression of c-IAP1 protein in gastric adenocarcinoma was significantly higher than that of normal gastric mucosa (P<0.05);The expression of c-IAP1 protein in gastric adenocarcinoma tissues were significantly correlated with histological differentiation,TNM clinical stage, lymph node metastasis and depth of invasion (P<0.05),while there was no correlation with gender and age (P>0.05). There was a negative correlation between the expression of Caspase-9 and c-IAP1 in gastric adenocarcinoma tissues (r s=-0.578, P<0.05). Conclusion The expression of Caspase-9 and c-IAP1 in gastric adenocarcinoma tissues was negatively correlated, and their abnormal expression participated in the development and progression of gastric adenocarcinoma.

EXPRESSION AND SIGNIFICANCE OF APOPTOSIS RELATED FACTORS CCAR1 AND PAR-4 IN GASTRIC ADENOCARCINOMA

Objective: To investigate the expression and significance of cell division cycle and apoptosis regulator protein1 (CCAR1) and prostate apoptosis response-4 gene (Par-4) protein in gastric adenocarcinoma. Methods: SP immunohistochemical staining was used to detect the expression of CCAR1 and Par-4 in 60 gastric adenocarcinoma tissues and 40 paracancerous normal gastric mucosa tissues. The relationships between CCAR1, Par-4 and clinicopathological parameters of gastric adenocarcinoma, as well as the correlation of CCAR1 and Par-4 in gastric adenocarcinoma were analyzed. Results: The positive expression rate of CCAR1 protein in gastric adenocarcinoma tissues was significantly higher, while the Par-4 protein was significantly lower than that of normal gastric mucosa (P<0.05). The CCAR1 protein expression in gastric adenocarcinoma was related to differentiation degree (P<0.05); The expression of par-4 protein was related to differentiation degree, TNM stage and lymph node metastasisa (P<0.05). Spearman correlation analysis showed that no correlation between the expression of CCAR1 protein and Par-4 protein in gastric adenocarcinoma (r=-0.08, P>0.05). Conclusions: Par-4 and CCAR1 involve in the occurrence and progression of gastric adenocarcinoma together, but they may be regulated by different mechanisms.

EXPRESSION AND SIGNIFICANCE OF INTEGRIN Α5Β1, HPA IN PSTT AND CHORIOCARCINOMA

Objective:To investigate the expression and significance of integrin α5β1 and heparanase (HPA) in placental site trophoblastic tumor (PSTT) and choriocarcinoma tissue. Methods:Western blot was used to detect the integrin α5β1 and HPA protein expression in 20 cases choriocarcinoma tissue, 15 cases PSTT tissue and 40 cases normal villi. Results:The integrin α5β1 and HPA protein expression in choriocarcinoma tissue were obviously higher than PSTT tissue and normal villi (P<0.05); The integrin α5β1 and HPA protein expression in PSTT tissue were obviously higher than normal villi (P<0.05). Conclusions:Abnormal expression of integrin α5β1 and HPA maybe closely related to the development of choriocarcinoma and PSTT.
