Analysis of the Relationship between Levels of VEGF, MMP-9, tPA and Pognosis in Patients with Endometriosis

Objective To explore the relationship between the prognosis and vascular endothelial growth factor (VEGF), matrix metalloproteinase-9 (MMP-9), tissue plasminogen activator (tPA) in patients with endometriosis (EM). Methods Eighty-four EM patients, who were treated with conservative surgery, were selected as the study objects. According to the follow-up of 1 year after surgery, they were divided into recurrence group (11 cases) and non recurrence group (73 cases). The levels of VEGF, MMP-9 and tPA were measured before and after operation, and ROC curve was used to analyze the prognostic value of VEGF, MMP-9 and tPA in EM patients. Results Before surgery, there was no significant difference in the levels of VEGF, MMP-9, and tPA between the two groups of patients (P>0.05). On the 7th day after operation, the levels of VEGF and MMP-9 of the two groups were lower than that of preoperatively, while the levels of tPA were higher than that of preoperative, the differences were statistically significant (P<0.05). The levels of VEGF and MMP-9 were lower than that of patients in the recurrence group, and the tPA level was higher than that of patients in the recurrence group, the difference were statistically significant (P<0.05). ROC curve analysis showed that the AUC of serum VEGF, MMP-9, and tPA levels in evaluating the prognosis of EM patients were 0.783, 0.766, and 0.788, respectively, which had certain accuracy, and the evaluation value of the three-item combination (AUC=0.808, P<0.01) was higher. Conclusion The levels of VEGF, MMP-9, and tPA in patients with endometriosis are closely related to the prognosis, and the three combined tests are more valuable for their prognostic evaluation.

The Sensitivity, Specificity and Accuracy of Color Doppler Ultrasound in Diagnosing Deep Venous Thrombosis of Lower Limbs

Objective To investigate the sensitivity, specificity and accuracy of color Doppler ultrasound in the diagnosis of lower limb deep vein thrombosis. Methods The clinical data of 88 patients with lower limb disorders who had admitted to our hospital from January 2018 to January 2019 were retrospectively analyzed. All the patients were diagnosed by color Doppler ultrasound. The sensitivity, specificity and accuracy of color Doppler ultrasound in the diagnosis of lower limb deep vein thrombosis were evaluated by using digital subtraction angiography as the gold standard. Results Angiography showed that 60 cases of deep venous thrombosis and 28 cases of non-deep venous thrombosis were found in 88 patients with lower extremity disorders. According to the diagnosis of color Doppler ultrasound, 58 cases were lower extremity deep vein thrombosis patients and 30 cases were non-lower limb deep vein thrombosis patients. The accuracy rate was 95.45%, the sensitivity was 95.00%, and the specificity was 96.43%. The accuracy rate of color Doppler venous examination and angiography was 95.00% There was no significant difference in the results of shadow vein examination (χ 2 = 2.274, P = 0.132). Conclusion Color Doppler ultrasound has good diagnostic efficacy in diagnosing patients with deep venous thrombosis of lower limbs, which is beneficial to provide reference for clinical diagnosis and treatment.

EFFECTS OF SERICIN ON PI3K MRNA EXPRESSION IN PANCREAS OF TYPE 2 DIABETES MELLITUS RATS

Objective: To investigate the effects of sericin on phosphatidylinositol-3-kinase (PI3K) mRNA expression in pancreas of type 2 diabetic rats. Methods: Male SD rats were randomly divided into three groups: normal control group, diabetic model group and sericin treatment group. The rat model of type 2 diabetes mellitus was established by feeding high-fat and high-sugar diet and intraperitoneal injection of streptozotocin (35mg/kg, twice). Then the rats in the sericin treatment group were given sericin (2.4g/kg/d) by gavage. Glucose oxidase method was used to detect the blood glucose of rats, and Real Time Q-PCR were used to detect the expression of PI3K mRNA in pancreas of rats in each group. Results: Compared with the normal control group, the blood glucose of diabetic rats increased significantly and the expression of PI3K mRNA in pancreas decreased significantly (P<0.05); Compared with diabetic rats, the blood glucose of sericin treated rats decreased significantly, and the expression of PI3K mRNA in pancreas increased significantly (P<0.05). Conclusions: Sericin may regulate insulin PI3K-Akt signaling pathway by increasing the expression of PI3K in pancreas, thus reducing blood glucose .

The type 2 diabetic rats' model was established by high fat and high sugar feed combined streptozotocin injection. The rats in sericin medication group were lavaged with sericin (2.4g/kg) after the diabetic model was successfully established. Glucose oxidase method was used to detect the blood glucose of rats, Real Time Q-PCR to detect the IRS-1 mRNA expression in pancreas.ResultsCompared with rats in normal control group, the blood glucose of rats in diabetic model group increased obviously, the IRS-1 mRNA expression in pancreas decreased obviously (P<0.05). Compared with rats in diabetic model group, the blood glucose of rats in sericin medication group decreased significantly, the IRS-1 mRNA expression in pancreas increased significantly (P<0.05).ConclusionsSericin may reduce blood glucose by up regulating the expression of IRS-1 in pancreas.

EFFECTS OF SERICIN ON EXPRESSION OF BCL-2 PROTEIN IN RATS’ INS-1 CELLS

Objective: To investigate the effects of sericin on expression of Bcl-2 protein in rats’ INS-1 cells. Methods: INS-1 cells were divided into normal control group, streptozotocin (STZ)-injuried group and sericin-treatment group. The INS-1 cells in normal control group were cultured with complete medium without any medicine; the INS-1 cells in STZ-injuried group were cultured with complete medium containing 10mmol/L STZ; the INS-1 cells in sericin-treatment group were cultured with complete medium containing 10mmol/L STZ and 300μg/ml sericin. Western Blotting was used to detect the expression of Bcl-2 protein in INS-1 cells of each group. Results: Compared with normal control group, the Bcl-2 protein expression in INS-1cells of STZ-injuried group decreased obviously (P<; 0.05). Compared with STZ-injuried group, the Bcl-2 protein expression in INS-1 cells of sericin-treatment group increased obviously (P<; 0.05). Conclusions: Sericin may inhibit beta cell apoptosis by up regulating the expression of Bcl-2 protein.
