Objective
To detect the expression of myelodysplastic leukemia factor 1-interacting protein (MLF1IP), naked cuticle homolog protein 1 (NKD1), and Golgi phosphoprotein 3 (GOLPH3) in gastric cancer tissues, and to determine the relationship between them and the level of immune cell infiltration and drug sensitivity.
Methods
The research subjects were the cancer and adjacent tissues of 51 patients with gastric cancer who were treated at the First People's Hospital of Shangqiu City from January 2022 to January 2024. The expressions of GOLPH3, MLF1IP and NKD1 were detected by immunohistochemistry. Analyze the relationship between the expression of MLF1IP, NKD1 and GOLPH3 and the infiltration level of immune cells as well as the sensitivity to different drugs.
Results
Compared with adjacent tissues, the expression of GOLPH3, the level of macrophages, the expression of NKD1 and the expression of MLF1IP in gastric cancer tissues were higher, while the levels of CD8
+
T cells, B cells, CD4
+
T cells, neutrophils and dendritic cells were lower. The differences were statistically significant (P<0.05). The drug resistance of doxorubicin (ADM) and cisplatin (DDP) in gastric cancer tissues was higher than that of fluorouracil (5-FU) and mitomycin (MMC), and the difference was statistically significant (P<0.05). NKD1, MLF1IP and GOLPH3 were negatively correlated with CD8
+
T cells, neutrophils, B cells, CD4+T cells and dendritic cells, and positively correlated with macrophages. Compared with the negative expressions of NKD1 and MLF1IP, the positive expressions of NKD1 and MLF1IP have higher drug resistance and lower sensitivity for MMC, DDP, 5-FU and ADM. Compared with the negative expression of GOLPH3, the positive expression of GOLPH3 had higher sensitivity and lower drug resistance in ADM, 5-FU, DDP and MMC, and the difference was statistically significant (P<0.05).
Conclusion
MLF1IP, NKD1 and GOLPH3 are highly expressed in gastric cancer tissues, closely related to the sensitivity of chemotherapy drugs, and can inhibit the infiltration of immune cells.

Objective
To investigate the sensitization effect of Ailanthone (AIL) on cisplatin-resistant A549/DDP cells of non-small cell lung cancer.
Methods
The effects of AIL on the viability of A549 and A549/DDP cells were detected by MTT. Analysis of the combined drug use index(CI) of Ailanone and cisplatin using Chou-Talalay. Cell cycle and apoptosis were detected by flow cytometry, and the expression of apoptosis-related proteins were detected by Western blotting.
Results
AIL (0.6μmol/L) and DDP (50μg/mL) had synergistic effect. Cell cycle arrest and apoptosis rate increased in G1 phase. The protein expression of caspase-3 and bcl-2 decreased, but the protein expression of Cleaved Caspase3, Bax, CDK4 and CyclinD1 increased.
Conclusion
AIL enhanced the sensitivity of DDP to A549/DDP, and increased the growth inhibition and apoptosis of DDP on A549/DDP.

Objective:To analyze the diagnostic value of serum procalcitonin (PCT) detection in children with bacterial upper respiratory tract infection. Methods:The serum PCT and C reactive protein (CRP) level, white blood cell (WBC) and sputum culture of 108 children with suspected bacterial upper respiratory tract infection that were treated in Qingdao Eighth People's Hospital from 2015.7 to 2017.5 were detected. The diagnostic value of serum PCT level,CRP and WBC for children with bacterial upper respiratory tract infection were analyzed by taking the results of sputum culture as gold standard. Results:Among 108 children, the sputum culture showed 72 children with bacterial upper respiratory tract infection. The serum PCT, CRP level and WBC of children with bacterial upper respiratory tract infection were all obviously higher than non bacterial upper respiratory tract infection (P<0.05). Taking the results of sputum culture as gold standard, the sensitivity, specificity, positive predictive value, negative predictive value and accuracy rate of serum PCT in diagnosis of bacterial upper respiratory tract infection were higher than CRP and WBC (P<0.05). Conclusions:The serum PCT detection has higher diagnostic value for children with bacterial upper respiratory tract infection, and has higher diagnostic sensitivity and specificity than CRP and WBC.
