Effects of Overexpression of Tmod1 on Migration and Invasion of Vascular Smooth Muscle Cells

Objective To investigate the effect of tropomodulin 1 (Tmod1) overexpression on the migration and invasion of rat vascular smooth muscle cells (VSMCs). Methods Rat vascular smooth muscle cells were cultured and identified by immunofluorescence SM22a staining. Adenovirus encoding Tmod1 was used to infect VSMCs and control adenovirus Ad-null,was used to infect control VSMCs. After 48 hours of culture, the expression of Tmod1 in each group was detected by Western blot to identify the overexpression efficiency. The cell scratch test and Transwell assay can detect the fuction of cell migration. Rt-qpcr was used to detect the expression of matrix metalloproteinase MMP15 after Tmod1 overexpression. Results Overexpression of Tmod1 promoted the migration and invasion of rat vascular smooth muscle cells (P<0.05), and the expression of MMP15 was significantly increased (P<0.05). Conclusion Tmod1 overexpression may promote cell migration and invasion possibly by targeting MMP15 expression.

The Role of Tropomyosin 1 and CDK1 in the Regulation of Body Water

Objective To analyze the role of tropomodulin 1 (Tmod1) in the regulation of body water. Methods Using mouse kidney primary inner medullary collecting duct (IMCD) cells for the study, the primary cells were transfected with adenovirus to overexpress/knock down Tmod1. Western blot and real-time fluorescent quantitative PCR were used to detect the changes in Tmod1 protein and RNA levels to verify adenovirus efficiency. Subsequently, the expression of cyclin-dependent kinase 1 after overexpression/knockdown of TMOD1 was detected by Western blot. Results After the overexpression of Tmod1, the level of CDK1 protein increased significantly, and decreased after knockdown of TMOD1, with statistical significance (P<0.05). Conclusion Tmod1 may participate in the process of body water mediation by influencing CDK1, but its mechanism remains to be further studied.

Objective: To investigate the regulatory effects of matrix stiffness on the elastic modulus and cytoskeleton of macrophages. Methods: Mouse macrophage RAW264.7 were cultured on different stiffnesses of polyacrylamide gels (2kPa and 20kPa). The nano-indenter was used to detect the elastic modulus of macrophages cultured on different matrix stiffness gels, flow cytometry to detect the F-actin content in macrophages. Results: Compared with 2kPa matrigel, elastic modulus and F-actin content of macrophages that cultured on 20kPa matrigel increased obviously (P<0.05). Conclusions: Matrix stiffness can regulate the elastic modulus and cytoskeleton of macrophages, but the mechanism needs to be further studied.
