过表达LC3基因对横纹肌肉瘤RD细胞生物学行为的影响

Effect of Overexpression of LC3 Gene on the Biological Behavior of Rhabdomyosarcoma RD Cells

摘要: 目的 探究过表达LC3基因介导的自噬对横纹肌肉瘤(RMS)RD细胞生物学作用的影响。方法 采用转染技术将RD细胞分为阴性对照组(转染空载质粒pcDNA3.1)和LC3实验组(转染LC3质粒)。通过RT-qPCR技术及Western Blot技术验证转染LC3基因在mRNA水平及蛋白质水平的表达变化;采用CCK-8实验分析RD细胞增殖情况;运用细胞划痕愈合实验及Transwell实验检测细胞迁移和侵袭能力;使用TUNEL实验评估细胞凋亡水平。结果 与阴性对照组相比,LC3实验组在RD细胞中的mRNA表达量显著升高(P<0.001);其过表达可抑制RD细胞的增殖、迁移、侵袭(均P<0.05),并促进其细胞凋亡(P<0.001)。结论 LC3基因可抑制RD细胞增殖、迁移、侵袭能力并促进细胞凋亡,有望成为治疗RMS新的靶基因。

Abstract: Objective This study explored the effect of autophagy mediated by overexpression of the LC3 gene on the biological functions of rhabdomyosarcoma RD cells. Methods Rhabdomyosarcoma RD cells were divided into a negative control group (transfected with empty vector pcDNA3.1) and an LC3 experimental group (transfected with LC3 plasmid) using transfection technology. The mRNA and protein expression levels of the transfected LC3 gene were detected using RT-qPCR and Western blot assays. Cell proliferation was analyzed by the CCK-8 assay, while cell migration and invasion abilities were evaluated using wound healing and Transwell assays. Cell apoptosis was assessed by the TUNEL assay. Results Compared with the negative control group, the mRNA expression level was significantly increased in rhabdomyosarcoma RD cells in the LC3 experimental group (P<0.001). In addition, LC3 overexpression inhibited the proliferation, migration, and invasion of RD cells (all P<0.05), and promoted their apoptosis (P<0.001). Conclusion The LC3 gene inhibits the proliferation, migration, and invasion ability of RD cells and promotes cell apoptosis, and it is expected to become a novel target gene for the treatment of rhabdomyosarcoma.
